中药穴位贴敷对急性气管-支气管炎模型大鼠的保护作用研究

摘　　要：目的 研究穴位贴敷对烟熏所致急性气管-支气管炎大鼠外周血及肺组织肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)水平以及支气管上皮细胞核转录因子-κB(NF-κB) p65表达的影响。方法 采用烟熏法制备急性气管-支气管炎模型大鼠,贴敷组用中药粉(麻黄、杏仁、黄芩)+消肿止痛贴于双侧肺俞穴贴敷,西药组灌胃头孢克洛。治疗7 d后,以光镜观察组织病理学,ELISA法检测血清及肺组织TNF-α、IL-1β水平,免疫组化法检测NF-κB p65表达。结果 穴位贴敷显著降低血清及肺组织TNF-α、IL-1β水平,抑制支气管上皮细胞NF-κB p65表达,减轻支气管以及肺组织的损伤。结论 穴位贴敷可显著减轻烟熏引起的急性气管-支气管炎,保护机制可能与调控NF-κB p65表达以及TNF-α、IL-1β水平相关。Objective:To study the effect of acupoint application on the levels of TNF-α,and IL-1βin periph-eral blood and lung tissue and the expression of NF-kB p65 in bronchial epithelial cells of rats with acute bronchi-tis-tracheitis induced by smoking.Methods:After acute bronchitis-tracheitis rats were modeled by smoking ex-pouse,the acupoint application group was used acupoint application on bilateral Feishu points with TCM drugs powder and herb liquor.The western group was used cafaclor orally.The treatment period was lasted for 7 days.Pathological changes of the bronchus and lung were observed under the light microscope,ELISA detected the lev-els of TNF-αand IL-1βin serum and lung tissue,and the expression of NF-kB p65 was detected by immunohis-tochemistry.Results:Acupoint application significantly reduced the levels of TNF-αand IL-1βin serum and lung tissue,inhibited the expression of NF-kB p65 in bronchial epithelial cells,and alleviated the damage of bron-chus and lung tissue.Conclusion:Acupoint application can significantly reduce acute bronchitis-tracheitis caused by smoking.The protective mechanism may be related to regulating NF-kB p65 expression and TNF-αand IL-1βlevels.
