供试品制备方式对气相色谱法测定人血白蛋白辛酸钠含量影响的研究

摘　　要：目的:验证两种供试品制备方式对气相色谱法测定人血白蛋白辛酸钠含量结果的影响,为提高辛酸钠含量测定的准确性、规范辛酸钠含量测定方法提供参考依据。方法:参照《中华人民共和国药典》2020年版通则中“3111辛酸钠测定法”进行辛酸钠含量测定,但加入三氯甲烷离心后不进行挥干处理,直接注入1μL于气相色谱仪中进行分析[色谱条件:色谱柱HP-FFAP(0.53 mm×30 m×1.0μm);气化室温度230℃;色谱柱温度160℃;检测器温度230℃;载气为氮气;柱流速为15.29 mL/min;进样模式为不分流]。分别采用加1.5 mol/L高氯酸溶液之后进行混合的方式和在振荡器上边混合边加1.5 mol/L高氯酸溶液的方式进行供试品处理,并对试验结果进行对比研究。结果:加1.5 mol/L高氯酸溶液之后进行混合的供试品在离心后会偶然产生凝块,且检测结果偏小;在振荡器上边混合边加1.5 mol/L高氯酸溶液的供试品离心后不会产生凝块,且检测结果稳定。结论:采用气相色谱法测定人血白蛋白辛酸钠含量供试品制备中加1.5 mol/L高氯酸溶液方式的不同对辛酸钠含量结果的影响比较明显,在进行人血白蛋白供试品制备时在振荡器上边混合边加1.5 mol/L高氯酸溶液可保证检测结果的准确性。Objective:To verify the effects of two preparation methods on the determination of sodium caprylate in human blood by gas chromatography,and to provide reference for improving the accuracy of the determination of sodium caprylate and standardizing the determination method of sodium caprylate.Methods:The content of sodium caprylate was determined according to the“3111 sodium caprylate determination method”in the General rules of the People’s Republic of China Pharmacopoeia(2020 edition).However,after centrifuging with trichloromethane,it was directly injected 1μL into the gas chromatograph for analysis.The chromatographic column was HP-FFAP(0.53 mm×30 m×1.0μm).Gasification chamber temperature 230℃;Column temperature 160℃;Detector temperature 230℃;The carrier gas is nitrogen;The column flow rate was 15.29 mL/min.The injection mode is no shunt].The samples were treated by adding 1.5 mol/L perchloric acid solution and adding 1.5 mol/L perchloric acid solution on the mixing side of the oscillator,and the test results were compared.Results:The samples mixed with 1.5 mol/L perchloric acid solution would occasionally produce clots after centrifugation,and the detection results were small.The test product with 1.5 mol/L perchloric acid solution added on the mixing side of the oscillator did not produce clots after centrifugation,and the test result was stable.Conclusion:The different ways of adding 1.5mol/L perchloric acid solution in the preparation of human blood albumin sodium caprylate by gas chromatography have obvious influence on the results of sodium caprylate content.Adding 1.5mol/L perchloric acid solution on the mixing side of the mixer can ensure the accuracy of the detection results.
