摘 要:目的 明确通络熄风汤改善糖尿病心肌损伤大鼠的治疗效果及机制。方法 本实验将大鼠分为正常组、模型组以及治疗组,利用STZ注射法将大鼠统一构建为糖尿病模型。治疗组予以中药配方颗粒通络熄风汤悬浮液灌胃治疗。采用Western blotting方法检测心肌细胞中P62、Mst1、Sirt3蛋白的含量,运用HE染色观察大鼠心肌组织结构变化,以评价通络熄风汤对糖尿病心肌损伤改善效果及机制。结果 与模型组相比,治疗组显著改善糖尿病大鼠的体重及血糖指标,差异有统计学意义(P<0.01)。同时与模型组相比,通络熄风汤治疗后大鼠心肌细胞的排列情况与坏死状态均有明显改善。与模型组相比,治疗组大鼠心肌组织中的Mst1、P62蛋白含量明显减少,Sirt3蛋白含量明显提高,差异均具有统计学意义(P<0.01)。结论 通络熄风汤可以对心肌组织细胞自噬水平进行调整,并保护心脏功能,上述机制通过Mst1通路发挥作用。Objective To clarify the therapeutic efficacy and mechanism of Tongluo Xifeng Tang in relieving diabetic myocardial injury in rats.Methods All rats were divided into normal group,model group and treatment group,and diabetic model was established by using STZ injection method.The treatment group was intragastrically given suspension of granules of Tongluo Xifeng Tang.The content of P62,Mstl and Sirt3 proteins in cardiomyocytes were detected by using Western blotting assay.The changes of myocardial tissue structure were observed by using HE staining for reviewing the efficacy and mechanism in relieving diabetic myocardial injury in rats.Results Compared with model group,the body weight and blood glucose indexes were significantly improved in treatment group(P<0.01).Compared with model group,the arrangement and necrotic state of cardiomyocytes were significantly improved in treatment group.Compared with model group,the content of Mstl and P62 proteins decreased significantly,and content of Sirt3 protein increased significantly in treatment group(P<0.01).Conclusions Tongluo Xifeng Tang can adjust the autophagy level of myocardial tissue cells and protect heart function.The above-mentioned mechanism works through the Mst1 pathway.
