血清miR-935和miR-22-3p水平与小儿支气管哮喘严重程度的相关性分析

摘　　要：目的探讨分析小儿支气管哮喘患者发作时血清miR-935和miR-22-3p的水平变化及其与病情严重程度的相关性。方法将观察组50例支气管哮喘患儿根据病情严重程度分为轻度组(n=17)、中度组(n=18)和重度组(n=15),同期50例来院健康体检儿童作为对照组,采集外周血进行血清miR-935和miR-22-3p水平检测,比较血清miR-935和miR-22-3p在观察组、对照组及观察组不同病情患儿间的水平表达差异并分析其对儿童支气管哮喘的诊断价值。结果观察组患儿血清miR-935表达水平明显高于对照组,而血清miR-22-3p表达水平呈相反趋势(P<0.05);轻度组、中度组和重度组患儿血清miR-935表达水平呈递增趋势,而血清miR-22-3p表达水平呈相反趋势(P<0.05);血清miR-935、miR-22-3p及两者联合对儿童支气管哮喘的诊断曲线下面积(AUC)分别为0.772、0.785和0.825,诊断灵敏度为72.25%、75.00%和82.25%,诊断特异性为75.00%、77.25%和85.00%。结论血清miR-935在支气管哮喘患儿中表达升高,而血清miR-22-3p表达降低,其水平的变化与疾病严重程度相关,对支气管哮喘具有一定的诊断价值。Objective To investigate the changes of serum miR-935 and miR-22-3p levels in children with bronchial asthma and their correlation with the severity of the disease.Methods 50 children with bronchial asthma in the observation group according to the severity of the disease were divided into mild group(n=17),moderate group(n=18)and severe group(n=15),while 50 children who came to the hospital for physical examination were taken as the control group.Peripheral blood was collected to detect the levels of miR-935 and miR-22-3p in serum,where the levels of miR-935 and miR-22-3p in serum were compared between the observation group and the control group,which of the differences of the level expression between children with different diseases and its diagnostic value in children with bronchial asthma were analyzed.Results The levels of serum miR-935 in the observation group were significantly higher than those in the control group,while the expression levels of miR-22-3p in serum showed an opposite trend(P<0.05).The expression levels of miR-935 in serum of children in mild,moderate and severe groups showed an increasing trend,while the expression levels of miR-22-3p in serum showed an opposite trend(P<0.05).The area under the diagnostic curve(AUC)of serum miR-935,miR-22-3p and their combination for bronchial asthma in children were 0.772,0.785 and 0.825,respectively,the diagnostic sensitivity was 72.25%,75.00%and 82.25%,and the diagnostic specificity was 75.00%,77.25%and 85.00%.Conclusion The expression of serum miR-935 is increased in children with bronchial asthma,hile the expression of serum miR-22-3p is decreased.The change of its level is related to the severity of the disease,which has certain diagnostic value for bronchial asthma.
