摘 要:目的探究原癌蛋白Fos(oncogene Fos,c-Fos)及丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)分子在多囊卵巢综合征(PCOS)并发子宫内膜癌患者中的表达,并通过对细胞周期的影响探究机制研究。方法选取2017年1月至2022年1月哈尔滨市阿城区人民医院妇科收集的28例PCOS并发子宫内膜癌患者子宫内膜癌组织作为试验组,以及功能失调性子宫出血患者部分子宫内膜组织作为对照组,通过qPCR对两组患者内膜c-Fos和MAPK mRNA表达水平进行检测。细胞学实验将人子宫内膜癌细胞HEC1B和Hela分别作为对照组和抑制剂组(MAPK抑制剂),采用CCK8法检测各组细胞增殖能力,流式细胞检测各组细胞的细胞存活、早期凋亡、晚期凋亡以及细胞周期G0/G1期、S期,G2/M期比例,qPCR和Western blot检测Fos、MAPK以及细胞周期相关分子G1/S-特异性周期蛋白-D1(G1/s-specific cyclin-D1,CyclinD1)和周期蛋白依赖性激酶1(cyclin-dependent kinases 1,CDK1)mRNA和蛋白的表达水平。结果组织学实验表明,相比于对照组子宫组织,试验组子宫内膜组织c-Fos和MAPK均显著高表达,差异有统计学意义(P<0.05)。细胞学实验表明:相较于对照组,抑制剂组的细胞增殖能力明显更低;细胞存活降低,早期凋亡和晚期凋亡增加,细胞周期G0/G1期显著增加,S期和G2/M期显著降低,差异有统计学意义(P<0.05),qPCR和Western blot实验表明抑制剂组细胞c-Fos、MAPK、CyclinD1、CDK1的mRNA和蛋白表达水平显著降低,差异有统计学意义(P<0.05)。结论c-Fos及MAPK分子在PCOS并发子宫内膜癌患者中呈高表达,其机制可能与细胞周期调控通路有关。Objective To investigate the expression of c-Fos and MAPK molecules in patients with polycystic ovary syndrome complicated with endometrial cancer,and to explore the mechanism by affecting the cell cycle.Methods The endometrial cancer tissues of 28 patients with polycystic ovary syndrome complicated with endometrial cancer collected from department of gynaecology Acheng People Hospital of Harbin from January 2017 to January 2022 were selected as the experimental group and endometrial tissues of patients with dysfunctional uterine bleeding were selected as the control group.The mRNA expression levels of oncoprotein Fos(c-Fos)and mitogen activated protein kinase(MAPK)were detected by qPCR.Two kinds of human endometrial cancer cells HEC1B and Hela were divided into control group and inhibitor group(MAPK inhibitor)respectively.The cell proliferation ability of each group was detected by CCK8 method,and the cell survival,early apoptosis,late apoptosis and cell cycle G0/G1 phase,S phase,G2/M phase were detected by flow cytometry.The expression levels of c-Fos,MAPK,G1/s-specific cyclin-D1(CyclinD1)and cyclin dependent kinases 1(CDK1)mRNA and protein were detected by qPCR and Western blot.Results The histological experiment showed that the expression of c-Fos and MAPK in endometrial tissue of the experimental group was significantly higher than that of the control group(P<0.05).Compared with the control group,the cell proliferation capacity was significantly lower in the inhibitor group.Cell survival decreased,early and late apoptosis increased,cell cycle G0/G1 phase increased significantly,S phase and G2/M phase decreased significantly,the difference was statistically significant(P<0.05).qPCR and Western blot showed that the mRNA and protein expression levels of c-Fos,MAPK,CyclinD1 and CDK1 in the inhibitor group decreased significantly(P<0.05).Conclusion c-Fos and MAPK are highly expressed in patients with polycystic ovary syndrome complicated with endometrial cancer,and its mechanism may be related to the regulation o
