摘 要:目的 建立不同种源麦冬高效液相色谱(HPLC)指纹图谱和HPLC同时测定麦冬二氢高异黄酮E、甲基麦冬黄烷酮A、甲基麦冬黄烷酮B的方法。方法 建立14批不同种源麦冬的HPLC指纹图谱,结合多元统计分析对不同种源麦冬进行质量评价并筛选出影响不同种源麦冬药材质量的差异性成分,采用HPLC法对质量差异性成分含量进行测定。结果 1)建立了不同种源麦冬的HPLC指纹图谱,标定共有峰23个,通过对照品比对指认其中5个共有峰,通过多元统计分析发现有10种成分是造成样品质量不同的差异性成分,其中3种成分分别为麦冬二氢高异黄酮E、甲基麦冬黄烷酮A、甲基麦冬黄烷酮B,这3种成分可以作为不同种源麦冬药材质量的差异性成分。2)建立了HPLC法同时测定麦冬二氢高异黄酮E、甲基麦冬黄烷酮A、甲基麦冬黄烷酮B的方法。结论 该研究所建立的方法操作简单,准确度高,可以用于不同种源麦冬定性定量研究,研究结果为川麦冬质量控制和资源评价提供依据。Objective To establish the HPLC fingerprints for Ophiopogonis Radix of different germplasm resources and an HPLC method for the simultaneous determination of ophiopogonanone E,methylophiopogonone A,and methylophiopogonone B.Methods HPLC fingerprints of 14 batches of Ophiopogonis Radix samples of different germplasm resources were established.On the basis of the fingerprints,multivariate statistical analysis was carried out to evaluate the quality of different samples and screen out the differential components that affect the quality of Ophiopogonis Radix.The content of the differential components was determined by the HPLC method.Results 1.The fingerprints of Ophiopogonis Radix of different germplasm resources were established,which showed 23 common peaks.After comparison with the reference substances,five common peaks of ophiopogonanone E,ophiopogonanone A,methylophiopogonone A,methylophiopogonone B,and ophiopogonanone C were identified.Ten components were identified as the differential components causing sample quality differences,in which ophiopogonanone E,methylophiopogonone A,and methylophiopogonone B can be used as the differential components to evaluate the quality of Ophiopogonis Radix of different germplasm resources.2.Furthermore,a HPLC method for the simultaneous determination of ophiopogonanone E,methylophiopogonone A,and methylophiopogonone B was established.Conclusion The method established in this study was simple and reliable and can be used for the qualitative and quantitative studies of Ophiopogonis Radix of different germplasm resources.The results provide a basis for the quality control and resource evaluation of Ophiopogonis Radix.
