摘 要:文章旨在研究复方中草药对运输后肉牛应激蛋白mRNA和蛋白表达水平的影响,试验将40头健康、体重相近的荷斯坦育肥牛随机分成4组,每组10个重复,分别为对照组(1组)、0.5%(2组)、1.0%(3组)、1.5%(4组)复方中草药添加组,饲喂14 d后模拟运输应激。采用ELISA测定运输前、后荷斯坦育肥牛血清中HSP 70含量,采用荧光定量PCR(qRT-PCR)和Western Blotting分别测定运输后组织中HSP 70、HSP 90及HSP 27的mRNA和蛋白质水平的表达量。结果显示,运输后的各组荷斯坦育肥牛血清中HSP 70含量均高于运输前的(P<0.05),与对照组相比,试验3、4组试验运输前和运输后荷斯坦育肥牛血清中HSP 70含量显著降低(P<0.05),试验3、4组运输后的肉牛肌肉组织HSP 27、HSP 70及HSP 90的mRNA的表达量显著降低(P<0.05);试验3、4组运输后的肉牛肌肉组织HSP 70和HSP 90蛋白表达量显著降低(P<0.05)。说明运输可以提高肉牛血清和肌肉组织中应激蛋白含量,添加不同水平的复方中草药可以降低运输后肉牛血清及肌肉组织中应激蛋白的水平,以添加量为1.0%较适宜。In order to study the effect of compound Chinese herbal medicine on the expression of stress protein mRNA and protein in beef cattle after transportation,40 healthy Holstein fattening cattle with similar body weight were randomly divided into 4 groups with 10 replicates in each group:control group(group 1),0.5%(group 2),1.0%(Group 3)and 1.5%(Group 4)compound Chinese herbal medicine adding group.After feeding for 14 days,transport stress was simulated.The content of HSP 70 in serum of Holstein fattening cattle before and after transportation was determined by ELISA.The mRNA and protein levels of HSP 27,HSP 70 and Hsp 90 in muscle tissue of Holstein fattening cattle after transportation were determined by QRT PCR and Western blotting,respectively.The results showed that the content of HSP 70 in serum of each group after transportation was higher than that before transportation(P<0.05).Compared with the control group,the content of HSP 70 in serum of test group 3 and 4 before and after transportation was significantly lower(P<0.05),and the content of HSP 27,HSP 70 and HSP 70 in muscle tissue of test group 3 and 4 after transportation were significantly lower(P<0.05)The mRNA expression of Hsp 90 was significantly decreased(P<0.05),and the protein expression of HSP 70 and Hsp 90 was significantly decreased(P<0.05).The results showed that transportation could increase the content of stress protein in serum and muscle tissue of beef cattle.The addition of different levels of compound Chinese herbal medicine could reduce the level of stress protein in serum and muscle tissue of beef cattle after transportation,and 1.0%was more suitable.
