摘  要:为了了解甘肃地区猪圆环病毒(PCV)的基因特征,试验采用PCR方法和基因测序技术,对甘肃省2018—2022年期间发生流行性腹泻的死亡仔猪病料进行回顾性检测,并对获得的毒株进行全基因测序,同时与参考毒株序列进行基因特征分析。结果表明:试验成功获得一株PCV-3毒株,将其命名为GSJN/2/2018毒株,该毒株的全基因组序列长度为1 999 nt,在1 149 nt处缺失碱基G,与参考毒株全基因序列的核苷酸相似性为97.8%~99.8%;ORF1和ORF2基因与参考毒株的核苷酸和氨基酸序列相似性分别为98.8%~99.9%/97.0%~100%和96.4%~99.7%/94.4%~100%;基于全基因组序列、ORF1和ORF2基因序列分别构建的系统进化树均将GSJN/2/2018毒株划分为PCV-3a基因型;该毒株主要线性抗原表位和3个滚环复制基序FTINN、HLQG、YCKK没有发生变异。说明通过回顾性检测获得的甘肃地区流行较早的GSJN/2/2018毒株属于PCV-3a基因型,并发现了新的基因缺失,丰富了PCV-3的基因组信息。In order to understand the genetic characteristics of Porcine circovirus(PCV) in Gansu Province,in this experiment,PCR method and gene sequencing technology were used to retrospectively detect the disease materials of piglet epidemic diarrhea in Gansu Province from 2018 to 2022.The complete gene of the obtained strain was sequenced,and the genetic characterization analysis was performed with the sequences of the reference strains.The results showed that a PCV-3 strain was successfully obtained in the experiment,which was named GSJN/2/2018 strain.The complete genome sequence of the strain was 1 999 nt in length,with a deletion of nucleotide G at the site 1 149 nt.The nucleotide homology with the complete gene sequences of the reference strains was 97.8%-99.8%.The nucleotide and amino acid homologies between ORF1 or ORF2 genes and the reference strains were 98.8%-99.9%/97.0%-100%,and 96.4%-99.7%/94.4%-100%,respectively.The phylogenetic trees were constructed based on the complete genome sequences,ORF1,and ORF2 gene sequences respectively,in which the GSJN/2/2018 strain was classified into the PCV-3a genotype.The main linear antigenic epitopes and three rolling circle replication FTINN、HLQG、YCKK of this strain didn't undergo mutations.The results suggested that through retrospective testing,the GSJN/2/2018 strain was obtained,which was found to be an earlier prevalent strain in Gansu Province,and belonged to the PCV-3a genotype;and new gene deletion had been discovered,enriching the genomic information of PCV-3.
