季节对各类型内蒙古白绒山羊瘤胃产甲烷菌多样性的影响

摘　　要：为研究不同季节对各个类型内蒙古白绒山羊瘤胃中产甲烷菌多样性的影响,试验选用阿尔巴斯型、二狼山型、阿拉善型内蒙古白绒山羊成年母羊各4头,在春(5月)、夏(8月)、秋(10月)、冬(2月)各季采集瘤胃液。采用高通量测序技术,通过特异性引物Met86f/Met1304r扩增瘤胃甲烷菌16S rDNA片段,建立克隆文库,并分析OUT类型、优势菌群等。结果表明:OTU-4型为冬季优势产甲烷菌;甲烷短杆菌属与甲烷球形菌属为白绒山羊瘤胃中优势菌群;春季、夏季、秋季、冬季的甲烷短杆菌属均值分别为93.05%、93.26%、75.67%、96.70%,甲烷球形菌属均值分别为6.95%、6.72%、4.92%、3.20%;按类型比较,二狼山型、阿尔巴斯型、阿拉善型山羊的甲烷短杆菌属均值分别为95.18%、92.60%、81.24%,甲烷球形菌属均值分别为2.59%、7.05%、6.71%。结果说明,不同季节不同类型的白绒山羊瘤胃中产甲烷菌组成有所不同,每个季节中甲烷短杆菌属与甲烷球形菌属都为白绒山羊瘤胃中优势菌群;但在秋季,白绒山羊瘤胃中产甲烷菌类型最丰富,其中二狼山型和阿拉善型瘤胃中的甲烷微菌属也是优势菌群。This study aimed to detect the effects of different seasons on the diversity of methanogens in the rumen of each types of Inner Mongolian white cashmere goats.In this experiment,we characterized the rumen methanogens of 3 different breed groups(Arbas,Erlangshan and Alashan Inner Mongolian white cashmere goats,n=4 for each group)in spring(May),summer(August),autumn(October)and winter(February).High-throughput sequencing technology thought specific primers Met86f/Met1304r amplify the page of rumen methanogens 16S rDNA and establish cione library to analysis the OUT types and dominant flora.The results showed that the OTU-4 type was dominant methanogen in winter,the Methanobrevibacter and the Methanosphaera were the dominant bacteria in the rumen of white cashmere goats.The mean value of Methanobrevibacter in spring,summer,autumn and winter was 93.05%,93.26%,75.67%,96.70%respectively,the mean value of Methanosphaera in each season was 6.95%,6.72%,4.92%,3.20%respectively.Different with seasons,the average of Methanobrevibacter in Erlangshan,Albas and Alashan was 95.18%,92.60%and 81.24%respectively;the average of Methanosphaera in Arbas,Alashan and Erlangshan was 7.05%,6.71%and 2.59%respectively.As a result of the white cashmere goats rumen methanogens composition was influenced by season or breed type.In each season,the Methanobrevibacter and the Methanosphaera were the dominant bacteria in white cashmere goats.The season with the most rumen methanogens in white cashmere goats was autumn,and the Methanomicrobium was also a dominant flora in rumen of Erlangshan and Alashan in autumn.
