摘 要:为提高发酵中草药中总黄酮的含量,为复方中草药发酵制备绿色饲料添加剂提供理论依据,本试验采用平板分离法,从自然堆积腐烂的中草药提取渣中分离到1株能发酵并提高复方中草药中总黄酮的芽孢杆菌,命名为py-3。采用形态学观察、生理生化试验并基于16S r DNA序列分析构建的系统发育树对菌株py-3进行了鉴定。通过单因素试验研究了菌株py-3固体发酵复方中草药条件对总黄酮含量的影响,并采用响应面试验对影响总黄酮含量较大的因素进行了优化。结果表明:菌株py-3被鉴定为贝莱斯芽孢杆菌。适宜的发酵条件为:葡萄糖6%,蛋白胨4%,KH2PO40.18%,MgSO4·7H2O 0.25%,Na Cl 3%。接种量15%,固水比1:1,发酵温度34℃,发酵时间6 d。此条件下,发酵物料中总黄酮含量可达6.163 mg/g,较发酵前总黄酮含量提高了68.99%。研究表明,贝莱斯芽孢杆菌py-3具有发酵复方中草药制备绿色饲料添加剂的应用前景。In order to improve the content of total flavonoids in fermented Chinese herbal medicines and provide theoretical basis for the preparation of green feed additives by fermentation of compound Chinese herbal medicines.A Bacillus strain named py-3,which could ferment and improve the total flavonoids in compound Chinese herbal medicine,was isolated from the natural accumulation and decay of Chinese herbal medicine extraction residue by plate separation method.The strain py-3 was identified by morphological observation,physiological and biochemical tests and phylogenetic tree constructed based on 16S rDNA sequence analysis.The effects of solid fermentation conditions of compound Chinese herbal medicine by strain py-3 on the content of total flavonoids were studied by using single factor experiment,and the factors affecting the content of total flavonoids were optimized by response test.The results showed that the strain py-3 was identified as Bacillus velezensis.The suitable fermentation conditions were as follows:glucose 6%,peptone 4%,KH2PO40.18%,MgSO4·7H2O 0.25%,NaCl 3%,inoculation amount 15%,solid water ratio 1:1,fermentation temperature 34℃,fermentation time 6 d.Under these conditions,the content of total flavonoids in the fermentation material reached 6.163 mg/g,which was 68.99%higher than that before fermentation.The study indicated that Bacillus velezensis py-3 had the application prospect of fermenting compound Chinese herbal medicine to prepare green feed additive.
