摘  要:甜菜盐胁迫下的转录组分析发现RNA解旋酶基因DExH17在盐胁迫下响应积极,本文利用生物信息学方法对甜菜RNA解旋酶基因DExH17的氨基酸序列基本特征和系统进化关系进行了预测和分析,利用实时荧光定量PCR方法分析了盐/干旱胁迫处理下甜菜DExH17基因的表达差异.结果表明:甜菜DExH17属于亲水性的不稳定蛋白,共有3个保守结构域,二级结构中无规则卷曲占比最大,不含有跨膜区,亚细胞定位于细胞核.通过DExH17氨基酸序列的比对分析发现甜菜与藜麦以及菠菜具有非常近的亲缘关系.定量分析发现,在对甜菜幼苗进行不同时长的盐/干旱胁迫处理过程中,DExH17基因的表达量有上调和下调两种不同程度的变化,推测DExH17基因响应盐/干旱胁迫,其可能参与调控甜菜的盐/干旱逆境响应过程.以上结果为抗盐/干旱性甜菜的培育提供科学依据.The transcriptome analysis of sugar beet under salt stress revealed that the RNA helicase gene DExH17 responded positively to salt stress.In this paper,the basic characteristics of the amino acid sequence and phylogenetic relationship of the RNA helicase gene DExH17 in sugar beet were predicted and analyzed using bioinformatics methods.The differential expressions of DExH17 gene in sugar beet under salt/drought stress were analyzed using fluorescence quantitative PCR method.The results showed that sugar beet DExH17 belongs to a hydrophilic unstable protein with three conserved domains,with the largest proportion of random coil in the sec-ondary structure and no transmembrane region.The subcellular localization is in the nucleus.Through the align-ment analysis of DExH17 amino acid sequence,it was found that sugar beet has a very close genetic relationship with quinoa and spinach.Quantitative analysis revealed that the expression level of DExH17 gene was up-regu-lated and down-regulated to varying degrees during salt/drought stress treatments of sugar beet seedlings.It is speculated that the DExH17 gene responds to salt/drought stress and may be involved in regulating the salt drought stress response process of sugar beet.The above results provide a scientific basis for the cultivation of salt/drought resistant sugar beet.
