摘 要:为了研究不同目数的材料对甜菜花粉的隔离效果,选择出最适合甜菜自交的隔离材料.以单胚保持系的糖甜菜和多胚的红甜菜为试验材料,分别设置了200目、250目以及300目共3种不同目数的隔离材料对单胚保持系的糖甜菜进行隔离,将多胚的红甜菜种植在外面,任自然风授粉,授粉后收集种子.将种子种植在发芽盒中,通过观察种子在发芽盒中生长的发芽率,同时结合分子标记的方法进行取样分析.结果表明:随着利用s17扩增的200目的隔离材料,1300 bp和1800 bp的条带最多,杂合率达到29%;300目的种子的隔离材料对甜菜花粉的隔离效果最好,杂合率为0;但随着目数的增多,甜菜自交罩中自交种子的发芽率也逐步降低,在不考虑发芽率的情况下,300目的材料隔离外来花粉的效果最好.In order to study the isolation effect of different mesh materials on beet pollen,the most suitable isola-tion materials for sugar beet self-inbreeding were selected.Taking the sugar beet of the monoembryo retention line and the red beet of the multi-embryo line as the test materials,three kinds of isolation materials with differ-ent mesh numbers of 200 mesh,250 mesh and 300 mesh were set up respectively to isolate the sugar beet of the single embryo retention line,and the multi-embryo red sugar beet was planted outside,pollinated by natural wind,and the seeds were collected after pollination.The seeds are planted in germination boxes,and the germi-nation rate of seeds growing in germination boxes is observed by observing the germination rate of seeds growing in germination boxes,combined with molecular markers.The results showed that with the 200 purpose isolation materials amplified by s17,the bands of 1300 bp and 1800 bp were the most,and the heterozygosity rate reached 29%.The isolation material of 300 seeds had the best isolation effect on beet pollen,and the heterozygosity rate was O;However,with the increase of mesh number,the germination rate of self-inbred seeds in the sugar beet inbred hood also gradually decreases,without considering the germination rate,300 mesh materials isolate for-eign pollen with the best effect.
