摘  要:目的 探讨洗涤对红细胞储存损伤的影响。方法 选取400 mL全血(保养液为ACD)制备的悬浮红细胞48袋,随机分成2组,每组24袋,1组不洗涤(对照组),1组于储存第14天洗涤,洗涤后的红细胞悬浮于MAP保养液中,分别于储存第d0、d7、d14、d21、d28、d35时检测对照组和洗涤组乳酸、K^(+)、Na^(+)、pH、游离血红蛋白(free hemoglobin, FHb)含量、红细胞渗透脆性和细胞膜完整性蛋白3(band-3 erythrocyte membrane integrity protein, band3),比较洗涤组和未洗涤组红细胞质量变化。结果 和对照组相比,洗涤组K^(+)、乳酸、PH值、游离血红蛋白下降(P<0.05),Na^(+)升高(d28、d35时,P<0.05),渗透脆性和band-3表达无差异。结论 对储存期的红细胞进行洗涤可以去除乳酸等有害代谢物质,改善储存环境,从而减少红细胞储存损伤,提高输血疗效。Objective To investigate the effect of washing on red blood cell storage lesion.Methods Toatlly 48 bags of suspended red blood cells prepared from 400 mL whole blood(maintenance solution as ACD)were selected and randomly divided into 2 groups with 24 bags each.The control group was not washed,the other group was washed on the 14th day of storage and suspended in MAP maintenance solution.Lactic acid,K^(+),Na^(+),pH,free hemoglobin content,erythrocyte osmotic fragility,band-3 erythrocyte membrane integrity protein were detected at do.d7、d14、d21,d28 and 35 of storage.The changes of erythrocyte between washed and unwashed groups were compared.Results Compared with the control group,K^(+),lactic acid,pH value and free hemoglobin in the washing group decreased(P<0.05),Na increased(at d28,d35,P<0.05),and there was no difference in the expression of band-3 and osmotic fragility.Conclusion Washing red blood cells during storage can remove harmful metabolites such as lactic acid,improve storage environment,reduce red blood cell storage damage and improve blood transfusion efficacy.
