摘  要:本研究采用ELISA和RT-PCR检测进口智利甜玉米种子(M118),ELISA检测玉米矮花叶病毒(maize dwarf mosaic virus,MDMV)的结果为阴性,RT-PCR扩增到635 bp的MDMV特异性片段,经测序证实该批种子携带MDMV。扩增并测定M118和已知MDMV阳性的智利甜玉米种子(M12)的外壳蛋白基因(coat protein,CP)序列,命名为MD118和MD12。CP序列分析结果显示,MD118和MD12分别含有873和912个碱基,对应的氨基酸为291和304个,二者核苷酸和氨基酸序列一致性分别为87.9%和92.8%。CP序列比对结果显示,MD118与匈牙利玉米分离物(FM883167)核苷酸序列一致性最高(97.6%),MD12与美国石茅分离物(MW026054,MW026053)核苷酸序列一致性最高(99.3%),表明这两个MDMV分离物可能来自智利不同地区。This study used ELISA and RT-PCR to detect the sweet corn seed lot(MI18)imported from Chile,and the ELISA result of maize dwarf mosaic virus(MDMV)was negative.The expected 635 bp amplicon of MDMV was obtained,and the determined sequence of the 635 bp amplicon confirmed that the seed lot was infected by MDMV.Complete coat protein(CP)genes of M118 and the positive corn seed sample M12 from Chile were amplified and sequenced,named MD118 and MD12.The CP sequence analysis results showed that MD118 and MD12 contained 873 and 912 bases,encoding 291 and 304 amino acids,respectively.The nucleotide sequence identity between the putative CP genes of MD118 and MD12 was 87.9%,and the amino acid sequence identity was 92.8%.The CP gene of MD118 was most closely related to the Hungarian corn isolate(FM883167)with the percent identity of 97.6%,while MD12 was most closely related to USA johnsongrass isolates(MW026054,MW026053)with identity of 99.3%.The CP sequence comparison results indicated that these two MDMV isolates might come from different regions in Chile.
