摘 要:为了探明河北石家庄地区5个貉养殖场发生肺炎死亡的原因,对病貉肺脏组织进行病原菌分离鉴定,共分离到5株大肠杆菌(DC1、DC2、DC3、DC4和DC5),并对分离菌的耐药性、耐药基因、致病性和毒力基因等进行研究。分离菌在MAC、SS和EMB培养基上长出菌落颜色分别为粉红色、红色和绿色金属光泽,革兰氏染色为革兰氏阴性短杆菌;生化试验结果符合大肠杆菌特性;16S rRNA测序结果与大肠杆菌序列的同源性超过99%;5株分离菌对青霉素、阿莫西林、四环素、甲硝唑和林可霉素的耐药率为100%;blaTEM和tetC耐药基因检出率均为100%,tetA检出率为80%;小鼠致病性试验结果显示,5株分离菌均可导致小鼠死亡,但小鼠死亡时间和只数范围存在差异;ECs3703和ECs3737毒力基因检出率为100%,iutA和iucD检出率为80%。研究表明,本次从石家庄地区分离到的貉大肠杆菌具有很强的耐药性和致病力,且携带有多种耐药基因和毒力基因。In order to find out the cause of death from pneumonia in five raccoon dog farms in Shijiazhuang,Hebei Province.The pathogen of lung tissue of diseased raccoon dogs was isolated and identified.Five strains of Escherichia coli were isolated and named DC1,DC2,DC3,DC4 and DC5 respectively.The drug resistance,drug resistance gene,pathogenicity and virulence gene of the isolated bacteria were studied.The results showed that the colony color of the isolated bacteria on MAC,SS and EMB medium was pink,red and green metallic luster respectively;Gram staining showed it was red negative bacilli;The biochemical test results were consistent with the characteristics of E.coli;The 16S rRNA sequences were more than 99% homologous to the known E.coli sequences;The resistance rate of 5 isolated bacteria to penicillin,amoxicillin,tetracycline,metronidazole and lincomycin was 100%;The detection rate of blaTEM and tetC was 100%,and the detection rate of tetA was 80%;The results of mouse pathogenicity test showed that the 5 strains of isolated bacteria could cause death of mice,but the time and number of mice died were different;The detection rate of ECs3703 and ECs3737 genes was 100%,and the detection rate of iutA and iucD genes was 80%.This shows that the E.coli isolated from Shijiazhuang has strong drug resistance and pathogenicity,and carries a variety of drug resistance genes and virulence genes.
