摘 要:本研究为完善南柴胡质量标准提供参考。对南柴胡进行实地采样,其中10批为栽培品,2批为野生品,通过基原鉴定、性状观察、检查和浸出物测定进行真伪优劣评价;建立显微鉴定、薄层色谱鉴别和含量测定,对采收期进行考察。结果显示,12批南柴胡为狭叶柴胡(Bupleurum scorzonerifolium)的干燥根,性状特征、水分、总灰分、酸不溶性灰分和浸出物测定结果符合《中国药典》(2020版)标准;新增加显微鉴别项,建立了薄层色谱鉴别方法,柴胡皂苷a(SSa)、柴胡皂苷c(SSc)和柴胡皂苷d(SSd)斑点清晰,分离度好,可用于南柴胡的专属性鉴定;建立了HPLC测定方法,SSa和SSd总量为0.02%~0.10%;以SSa和SSd含量为指标,参考根重,确定9月份为南柴胡适宜采收期。本研究完善了南柴胡药材的质量标准,为合理评价质量积累了必要的基础依据。This paper is to improve the quality standard of Bupleurum s corz one rifolium.Field sampling was conducted for B.scorzonerifolium,including 10 batches of cultivated products and 2 batches of wild products.The authenticity was evaluated through the identification of the original,the observation of the properties,the inspection and the determination of the extracts.The items of microscopic identification,thin-layer chromatography identification and content determination were established.Inspect the harvest period.The results showed that 12batches of Chinese herbal medicines were the dry roots of B.scorzonerifolium,and the determination results of properties,moisture,total ash,acid insoluble ash and extracts were in line with the standards of Chinese Pharmacopoeia(2020).A new microscopic identification item was added and a thin layer chromatographic identification method was established.The spots of saikosaponin a(SSa),saikosaponin c(SSc)and saikosaponin d(SSd) were clear and well separated,which could be used for the specific identification of B.scorzonerifolium.HPLC method was established.The total amount of SSa and SSd was 0.02%~0.10%.With SSa and SSd content as indexes and root weight as reference,September was determined as the suitable harvest time of B.scorzonerifolium.This study improved the quality standard of B.scorZonerifolium and accumulated the necessary experimental basis for reasonable the evaluation of quality.
