摘 要:目的探讨山苍子根提取物的遗传毒性及对大鼠的亚急性毒性效应。方法将山苍子树根按传统食用习惯提取,进行急性经口毒性试验、细菌回复突变试验、体外哺乳类细胞染色体畸变试验、哺乳动物红细胞微核试验和28 d经口毒性试验研究。结果山苍子根提取物LD50>60 g/kg·bw(相当于山苍子树根干重计500 g/kg·bw);细菌回复突变、体外哺乳类细胞染色体畸变、微核3项遗传毒性试验结果阴性,未观察到致突变性;亚急性毒性试验结果表明,在山苍子根提取物20.83、41.67和83.33 g/kg·bw剂量组中,个别剂量组大鼠的周体重、血液生化、血液学指标与水对照组比较,差异有统计学意义(P<0.05),但均在本实验室历史对照正常值范围内,其余各剂量组大鼠的体重、总进食量、总食物利用率、各器官的生长发育及组织形态等指标均未观察到明显的不良改变。结论山苍子根提取物为实际无毒级,未发现山苍子根提取物有遗传毒性作用,结合28 d经口毒性试验各指标统计结果综合分析,建议以5.0 g/kg·bw(相当于山苍子树根干重计41.66 g/kg·bw)作为未观察到有害作用剂量(NOAEL)。Objective To study the genetic toxicity of Litsea cubeba roots extract and its subacute toxic effects on rat.Methods The roots of Litsea cubeba were extracted and concentrated according to traditional eating habits for acute oral toxicity test,Ames test,in vitro mammalian cell chromosome aberration test,mammalian red blood cell micronucleus test,and subacute oral toxicity test.Results Acute oral toxicity test of Litsea cubeba roots extract LD_(50)60 g/kg·bw(Equivalent to Litsea cubeba roots dry weight 500 g/kg·bw),which was actually non-toxic according to the grading standard of acute toxicity dose.The result of genotoxicity tests(Ames test,in vitro mammalian cell chromosome aberration test,the mammalian red blood cell micronucleus test)were all negative and no mutations were obsered.In addition,the subacute toxicity test showed that in the 20.83,41.67,83.33 g/kg·bw dose groups,the weekly weight,blood biochemical and blood routine indicators of the individual dose groups were significantly different from those in the ultrapure water control group(P<0.05),but they are within the normal range of the historical control value of the laboratory,while no significant adverse changes were observed in the weight,total food intake,total food utilization,growth and development of various organs and histopathology of rats in the other dose groups.Conclusion In this study,Litsea cubeba roots extract was actually non-toxic,no genetic toxicity in Litsea cubeba roots extract were observed.Under the comprehensive analysis based on the statistical result of subacute oral toxicity test,5.0 g/kg·bw is recommended as NOAEL.
