摘 要:目的从生物力学的角度,观察褪黑素对去势小鼠椎体生物力学性能的影响。方法选取48只9周龄的雌性C57BL/6J小鼠,在无特定病原体(SPF)动物房适应性喂养3周至12周龄,到达性成熟后使用随机数字表法分为假手术组(Sham组,12只)和去卵巢组(36只);去卵巢组小鼠采取背侧切口行双侧卵巢切除术,而Sham组小鼠行双侧卵巢周围少量脂肪组织切除术。术后1周,将去卵巢组小鼠随机分为模型组(Control组)、己烯雌酚组(DES组)和褪黑素组(Melatonin组),每组12只。Melatonin组小鼠以褪黑素溶液灌胃,DES组小鼠以己烯雌酚溶液灌胃,Control组小鼠以等体积的2%乙醇溶液灌胃,Sham组以等体积的蒸馏水灌胃。所有小鼠经过不同方式干预后,分别于第4、8、12周末应用CO_(2)气体窒息方法,每组小鼠随机处死3只,完整取出各组小鼠的L3椎体,置于Instron万能材料试验机上进行生物力学测试。分析各组小鼠L3椎体最大压力负荷的变化,比较第4、8、12周末时各组小鼠L3椎体的最大压力负荷。结果各组小鼠L3椎体最大压力负荷随时间的变化均呈现下降的趋势,其中Sham组和DES组最大压力负荷高于Control组和Melatonin组,且曲线变化幅度稍小;Melatonin组最大压力负荷高于Control组。第4周末时,Melatonin组平均最大压力负荷为31.67 N,小于Sham组的35.67 N和DES组的36.67 N,但显著高于Control组的27.00 N,差异具有统计学意义(P<0.05)。第8周末时,Melatonin组平均最大压力负荷为28.00 N,小于Sham组的32.67 N和DES组的33.33 N,但显著高于Control组的23.33 N,差异具有统计学意义(P<0.05)。第12周末时,Melatonin组平均最大压力负荷为23.33 N,小于Sham组的29.00 N和DES组的29.33 N,但显著高于Control组的18.67 N,差异具有统计学意义(P<0.05)。结论褪黑素应用于去势的雌性C57BL/6J小鼠中,能够较为明显提高其L3椎体的最大压力负荷,从而起到减缓骨质疏松进程的作用。Objective To observe the effect of melatonin on the biomechanical properties of the vertebral body in ovariectomized mice.Methods 48 female C57BL/6J mice were selected at 9 weeks of age and fed in the specific pathogen free(SPF)animal house for 3 weeks to 12 weeks to reach sexual maturity,and then divided into a sham-operated group(Sham group,12 mice)and an ovariectomized group(36 mice)according to the random numerical table;the ovariectomized mice underwent bilateral ovariectomy with a dorsal incision,while the Sham group underwent bilateral excision of a small amount of adipose tissue around the ovaries.1 week after surgery,the ovariectomized mice were randomly divided into model group(Control group),diethylstilbestrol group(DES group),and Melatonin group,with 12 mice in each group.The mice in the Melatonin group were gavaged with melatonin solution,the mice in the DES group were gavaged with diethylstilbestrol solution,the mice in the Control group were gavaged with an equal volume of 2%ethanol solution,and the mice in the Sham group were gavaged with an equal volume of distilled water.All mice were randomly executed by CO2 gas asphyxiation at the end of the 4th,8th,and 12th week,respectively,and the L3 vertebrae of each group were removed intact and placed on an Instron universal material testing machine for biomechanical testing.The changes in the maximum pressure load on the L3 vertebrae of each group of mice were analyzed,and the maximum pressure load on the L3 vertebrae of each group of mice at the end of the 4th,8th,and 12th week were compared.Results The maximum pressure load of L3 vertebrae in all groups showed a decreasing trend with time,among which the maximum pressure load in Sham group and DES group was higher than that in Control group and Melatonin group,and the curve change was slightly smaller;the maximum pressure load in Melatonin group was higher than that in Control group.At the end of week 4,the mean maximum pressure load in the Melatonin group was 31.67 N,which was less than that of 35.67
