摘  要:为了探究盐胁迫下超氧化物歧化酶在绿豆中的作用,本研究对鉴定到的8个绿豆SOD基因进行生物信息学分析,并探究了盐胁迫下绿豆SOD基因的表达模式,结果表明,绿豆SOD蛋白都含有磷酸化位点,但磷酸化位点在数量和种类上都存在差异,说明它们可能被特定的蛋白激酶磷酸化;绿豆SOD蛋白大多定位在叶绿体中,部分存在于细胞质、过氧化物酶体、线粒体,说明它们主要在这些细胞器中清除活性氧;通过对SOD蛋白三级结构预测发现不同蛋白之间三级结构存在差异;通过系统进化分析,将SOD基因分为三个组;通过序列比对发现8个SOD基因可以分为Cu/Zn-SOD、Mn-SOD和Fe-SOD三种类型;研究发现绿豆8条SOD蛋白都可以响应盐胁迫,VrSOD2、VrSOD5、VrSOD6、VrSOD8在胁迫的各时间点表达都明显高于对照。本研究可以为盐胁迫下绿豆SOD基因功能研究提供参考。In order to explore the role of superoxide dismutase(SOD)in mung bean under salt stress,eight identified SOD genes in mung bean were analyzed by bioinformatics,and the expression pattern of SOD gene in mung bean under salt stress was explored.The results showed that there were phosphorylation sites in mung bean SOD protein,but there were differences in the number and types of phosphorylation sites,These results indicate that they may be phosphorylated by specific protein kinases;Most of the sod proteins in mung bean are located in chloroplast,some in cytoplasm,peroxisome and mitochondria,indicating that they mainly scavenge reactive oxygen species in these organelles;By predicting the tertiary structure of SOD protein,it was found that there were differences in the tertiary structure among different proteins;SOD genes were divided into three groups by phylogenetic analysis;Through sequence alignment,the eight SOD genes can be divided into three types:Cu/Zn-SOD,Mn-SOD and Fe-SOD;The results showed that 8 SOD proteins of mung bean could respond to salt stress,and the expression of vrsod2,vrsod5,vrsod6 and vrsod8 were significantly higher than that of the control at each time point of salt stress.This study can provide a reference for the study of SOD gene function in mung bean under salt stress.
