摘 要:本试验以铜陵白姜(Tongling white ginger)嫩芽为外植体,研究超声波处理时间和次氯酸钠处理时间、启动培养基、增殖培养基、壮苗及生根培养基以及炼苗方法对铜陵白姜快速繁殖体系建立的影响。结果表明,铜陵白姜外植体的最适处理为超声波处理30 min,次氯酸钠处理6 min,此时无菌率最高(83.33%)。铜陵白姜外植体适宜的萌发条件为MS+2.0 mg/L 6-BA+0.1 mg/L NAA+30 g/L蔗糖,萌发率为90.67%。铜陵白姜的适宜增殖培养基为MS+1.0 mg/L 6-BA+0.02 mg/L TDZ+0.10 mg/L NAA,增殖系数为5.67。GA_(3)为2.0 mg/L时,表现为苗粗壮,叶色深绿,长势好,适合壮苗培养。采用逐步炼苗法后,移栽至田间试验地成活率可达96.0%,该研究结果可为铜陵白姜的工厂化生产提供技术参数。Taking the tender buds of Tongling white ginger as explants,the effects of ultrasonic treatment time and sodium hypochlorite treatment time,starting medium,proliferation medium,strong seedling and rooting medium and seedling refining methods were studied on the establishment of rapid propagation system of Tongling white ginger.The results showed that the optimum treatment of Tongling white ginger explants was ultrasonic treatment for 30 min and sodium hypochlorite treatment for 6 min.at this time,the aseptic rate was the highest(83.33%).The suitable germination conditions of Tongling white ginger explants was MS+2.0 mg/L 6-BA+0.1 mg/L NAA+30 g/L sucrose,and the germination rate was 90.67%.The suitable proliferation medium of Tongling white ginger was MS+1.0 mg/L 6-BA+0.02 mg/L TDZ+0.10 mg/L NAA,and the proliferation coefficient was 5.67.When GA,was 2.0 mg/L,the seedlings were strong,dark green and grew well,which was suitable for strong seedling culture.The survival rate of transplanting to the field test site can reach 96.0%,and the results can provide technical parameters for the industrialized production of Tongling white ginger.
