摘 要:目的探讨畲药半边枫的不同溶剂提取物对类风湿关节炎大鼠的治疗作用。方法胶原诱导性关节炎大鼠模型造模采用尾根处2次皮下注射牛Ⅱ型胶原蛋白。选取56只雄性大鼠根据随机数字法分成7组,即对照组、CIA组、石油醚组、乙酸乙酯组、正丁醇组、水部位组和地塞米松组,每组各8只。各组大鼠连续30天灌胃半边枫不同溶剂提取物,评估各组关节炎指数和左足垫厚度,筛选治疗相关部位,同时测定各组胸腺、脾脏指数和踝关节Micro-CT成像。接着对关节滑膜进行病理染色和免疫组化染色,探讨其干预机制。结果半边枫各不溶剂提取物组与对照组比较,其中乙酸乙酯组大鼠左足垫厚度和关节炎指数降低最明显(P<0.05);与CIA组比较,乙酸乙酯组、地塞米松组胸腺指数和脾脏指数均降低,差异均有统计学意义(P<0.05)。Micro-CT成像结果显示,乙酸乙酯组和地塞米松组均能有效减轻CIA大鼠踝关节骨质破坏,对骨关节有保护作用。免疫组化结果显示,乙酸乙酯组对大鼠关节炎症的NF-κB和COX-2表达作用通路有抑制作用(P<0.05)。结论畲药半边枫乙酸乙酯提取物能有效减轻类风湿关节炎大鼠踝关节骨质破坏,具有良好的药用价值。Objective To explore therapeutic effect of SHE medicine Schefflera octophylla(Lour.)Harms extracts with different solvent on the treatment of rheumatoid arthritis(RA)rats.Methods Collagen-induced arthritis(CIA)rat model was made using twice subcutaneous injections of bovine typeⅡcollagen in the tail root after sufficient emulsification.56 rats were randomly divided into 7groups with random number:control group,CIA group,petroleum ether group,ethylacetate group,butanol group,water group and dexamethasone group,each with 8 rats.Rats in each group were continuously gavaged with different solvents of Schefflera Octophylla(Lour.)Harms for 30days.Joint index and left footpad thickness of each group were assessed to screen the treatment-related sites.Thymus index and spleen index and Micro-CT imaging of ankle joints were measured in each group.Pathological staining and immunohistochemistry staining of joint synovium were performed to explore the intervention mechanism.Results Comparing with control group,the footpad thickness and joint index of ethylacetate group rat were significantly decreased(P<0.05)in extracts with different solvent.Micro-CT results showed that the ethylacetate group and dexamethasone group could effectively reduce bone destruction in the ankle joints of model rats and had a protective effect.Immunohistochemical results showed that ethyl acetate group could inhibit the expression of NF-κB and COX-2 in rat joint inflammation(P<0.05).Conclusion SHE medicine Schefflera octophylla(Lour.)Harms extracts with ethylacetate could effectively reduce bone destruction in the ankle joints of rats with rheumatoid arthritis,and has medicinal benefits.
