摘  要:目的:探讨保和消积方对胃癌SGC-7901细胞增殖和迁移的影响,在mRNA和蛋白水平上阐明其分子机制。方法:选取对数生长期的胃癌SGC7901细胞作为研究对象,分为空白对照组和不同浓度(0.5、1、2、4、8、12和16 mg/mL)组,采用噻唑蓝(MTT)法检测不同时间(24、48、72 h)SGC7901细胞增殖情况;采用Transwell小室迁移试验检测SGC7901细胞的迁移能力;利用流式细胞仪检测SGC7901细胞凋亡情况;定量PCR和蛋白质免疫印迹法分别检测胃癌SGC7901细胞增殖、迁移、血管生成相关的磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT)、哺乳动物雷帕霉素靶蛋白(mTOR)、血管内皮生长因子(VEGF)mRNA和蛋白表达水平。结果:与空白对照组比较,作用不同时间后不同浓度的保和消积方组的胃癌SGC7901细胞增殖抑制率显著增加(均P<0.05),穿膜细胞数显著降低(均P<0.05),显著抑制了微管的形成(均P<0.05),AKT、mTOR、PI3K、VEGF mRNA和蛋白表达水平显著降低(均P<0.05)。结论:保和消积方通过抑制AKT、mTOR、PI3K、VEGF的mRNA和蛋白表达,从而抑制了SGC7901胃癌细胞增殖、迁移,促进了胃癌细胞的凋亡。Objective:To investigate the effect of Baohe Xiaoji Formula on the proliferation and migration of gastric cancer SGC-7901 cells and elucidate its molecular mechanism at the mRNA and protein levels.Methods:Gastric cancer SGC-7901 cells in the logarithmic growth phase were selected and divided into a blank control group and Baohe Xiaoji Formula groups at different concentrations(0.5,1,2,4,8,12,and 16 mg/mL).The MTT assay was used to detect cell proliferation at different time points(24,48,and 72 hours).The Transwell assay was performed to assess the migration ability of SGC-7901 cells.Flow cytometry was used to evaluate cell apoptosis.Quantitative PCR and Western blot were conducted to measure the mRNA and protein expression levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(Akt),mammalian target of rapamycin(mTOR),and vascular endothelial growth factor(VEGF)related to cell proliferation,migration,and angiogenesis in gastric cancer SGC-7901 cells.Results:Compared with the blank control group,the Baohe Xiaoji Formula groups showed increased proliferation inhibition rates of gastric cancer SGC-7901 cells after different durations of treatment(all P<0.05),reduced number of transmembrane cells(all P<0.05),inhibited tubule formation(all P<0.05),and decreased mRNA and protein expression levels of Akt,mTOR,PI3K,and VEGF(all P<0.05).Conclusion:Baohe Xiaoji Formula inhibits the proliferation and migration of SGC-7901 gastric cancer cells and promotes cell apoptosis by suppressing the mRNA and protein expression of Akt,mTOR,PI3K,and VEGF.
