摘  要:目的探讨miR-107在变应性鼻炎(AR)小鼠鼻黏膜中的表达及其作用机制。方法采用卵清蛋白(OVA)腹腔注射及滴鼻建立AR模型,实验分为Sham组(腹腔注射0.9%氯化钠溶液、滴鼻处理)、AR组、AR+miR-NC组(OVA致敏后用带有miR-NC的腺病毒注射入小鼠腹腔)、AR+miR-107组(OVA致敏后用带有miR-107过表达载体的腺病毒注射入小鼠腹腔),每组10只。qRT-PCR法检测外周血、鼻黏膜组织中miR-107表达量;采用视觉模拟评分量表(VAS)评估鼻部症状;试剂盒检测血清OVA特异性免疫球蛋白E(IgE)水平;ELISA法检测IL-4、IL-5、IFN-γ水平;Western blot检测NLRP3炎性小体相关蛋白(NLPR3、ASC)表达量。结果与Sham组比较,AR组外周血、鼻黏膜组织中miR-107表达水平、IFN-γ水平降低(P<0.05),鼻部症状评分、NLPR3、ASC蛋白水平升高(P<0.05),血清OVA特异性IgE、IL-4、IL-5水平升高(P<0.05);与AR组、AR+miR-NC组比较,AR+miR-107组外周血、鼻黏膜组织中miR-107表达水平、IFN-γ水平升高(P<0.05),鼻部症状评分、NLPR3、ASC蛋白水平降低(P<0.05),血清OVA特异性IgE、IL-4、IL-5水平降低(P<0.05)。结论AR小鼠中miR-107表达下调,miR-107过表达可抑制炎性反应、调节免疫功能,其作用机制与抑制NLRP3炎性小体活化有关。Objective To investigate the expression level of miR-107 in nasal mucosa of allergic rhinitis(AR)mice and its function.Methods AR model in mice was established by intraperitoneal injection and intranasal injection of ovalbumin(OVA),which was known as OVA sensitization.Mice were randomly divided into sham group(intraperitoneal injection of 0.9%NaCl solution and intranasal injection of OVA),AR group(OVA sensitization),AR+miR-NC group(OVA sensitization followed by intraperitoneal injection of adenovirus containing miR-NC),and AR+miR-107 group(OVA sensitization followed by intraperitoneal injection of adenovirus containing miR-107),with 10 mice in each group.The expression level of miR-107 in peripheral blood and nasal mucosa was detected by quantitative real-time PCR(qRT-PCR).Nasal symptoms were assessed using the Visual Analog Scale(VAS).The level of mouse OVA-specific immunoglobulin E(IgE)was measured by enzyme-linked immunosorbent assay(ELISA),as well as the levels of IL-4,IL-5 and IFN-γ.Western blot was used to detect the protein expressions of inflammasome-related proteins(NLPR3,ASC).Results Compared with those of the Sham group,the expression levels of miR-107 and IFN-γin peripheral blood and nasal mucosa tissues in AR group were significantly lower(P<0.05),while the nasal symptom score,the protein levels of NLPR3,and ASC,relative levels of OVA-specific IgE,IL-4 and IL-5 were significantly higher(P<0.05).Compared with those of AR group and AR+miR-NC group,the expression levels of the above indexes of AR+miR-107 group yielded the opposite results(all P<0.05).Conclusion MiR-107 is downregulated in AR mice,and its overexpression inhibits inflammation and regulates immune function in AR mice by suppressing NOD-like receptor protein 3(NLRP3)inflammasome activation.
