摘 要:目的:研究子痫前期胎盘组织中p57^(KIP2)基因表达水平与侵袭及凋亡的关系。方法:选择2020年3月—2022年9月南华大学附属第二医院收治的89例子痫前期患者作为研究组,其中55例轻度患者作为轻度子痫前期组、34例重度患者作为重度子痫前期组,另选取同期分娩的100例健康产妇作为正常对照组。收集三组胎盘组织,检测并比较p57^(KIP2)基因及侵袭、凋亡相关基因表达量的差异,采用Pearson检验分析子痫前期胎盘组织中p57^(KIP2)基因表达量与侵袭、凋亡相关基因表达量的相关性。结果:轻度子痫前期组、重度子痫前期组胎盘组织中p57^(KIP2)、G蛋白耦联受体30(GPR30)、胚层特异转录物同源物(MEST)、层粘连蛋白α4(LAMa4)、X-连锁凋亡抑制蛋白(XIAP)、生成素(Survivin)m RNA表达水平低于正常对照组,硫氧还蛋白互作蛋白(TXNIP)、含半胱氨酸的天冬氨酸蛋白水解酶-9(caspase-9)、Bcl2相关X蛋白(Bax)、Bcl2拮抗因子/杀伤(Bak)m RNA表达水平高于正常对照组,差异有统计学意义(P<0.05)。重度子痫前期组中上述基因m RNA表达水平变化均较轻度子痫前期组更为显著,差异有统计学意义(P<0.05)。子痫前期胎盘组织中p57^(KIP2)基因m RNA表达水平与侵袭相关基因、凋亡相关基因m RNA表达水平具有相关性(P<0.05)。结论:子痫前期胎盘组织中p57^(KIP2)基因表达水平降低与侵袭能力减弱、凋亡活性增强相关。Objective:To investigate the correlation of p57^(KIP2)gene expression and invasion,apoptosis in placenta of preeclampsia.Method:A total of 89 patients with preeclampsia who admitted to the Second Affiliated Hospital of Nanhua University from March 2020 and September 2022 were selected as the study group,including 55 mild patients as the mild preeclampsia group,34 severe patients as the severe preeclampsia group,and 100 healthy pregnant women who delivered at the same time were selected as the normal control group.The placental tissues of the three groups were collected to detect and compare the differences in the expression levels of p57^(KIP2)gene and genes related to invasion and apoptosis.Pearson test was used to analyze the correlation between the expression levels of p57^(KIP2)gene and the expression levels of genes related to invasion and apoptosis in placental tissues of preeclampsia.Result:The mRNA expression levels of p57^(KIP2),invasion related genes G protein-coupled receptor 30(GPR30),germ layer-specific transcript homolog(MEST),lamininα4(LAMa4),apoptosis related genes X-linked inhibitor of apoptosis protein(XIAP)and Survivin in the mild and severe preeclampsia groups were lower than those of the normal control group,the mRNA expression levels of invasion related gene thioredoxin interacting protein(TXNIP),cysteine-containing aspartate proteolytic enzyme-9(caspase-9),Bcl2-associated X protein(Bax),Bcl2 antagonist/killer(Bak)were higher than those in the normal control group,the differences were statistically significant(P<0.05).The changes in the expression of the above genes in the severe preeclampsia group were more significant than those in the mild preeclampsia group,the differences were statistically significant(P<0.05).The mRNA expression level of p57^(KIP2)gene in placenta tissue of preeclampsia is correlated with the mRNA expression levels of invasion related genes and apoptosis related genes(P<0.05).Conclusion:The decreased expression level of p57^(KIP2)gene in placenta tissue of preeclampsi
