摘　　要：目的:定性分析岩黄连总碱化学成分,并对其中8个含量较高的生物碱类成分进行定量分析。为研究岩黄连总碱的药效物质基础提供依据,也为岩黄连总碱质量控制提供参考。方法:采用超高效液相色谱-四级杆-飞行时间质谱联用技术(UPLC-Q-TOF-MS/MS)定性分析岩黄连总碱化学成分,并用高效液相色谱法(HPLC)同时测定岩黄连总碱中原阿片碱、非洲防己碱、延胡索乙素、黄连碱、脱氢卡维丁、巴马汀、小檗碱和白屈菜红碱的含量。结果:岩黄连总碱提取物中共鉴定出38个生物碱类成分,其中原小檗碱类13个,小檗碱类9个。此外,定量分析的8个成分在各自范围内线性良好(r>0.999 2),加样回收率96.70%~103.8%,RSD 0.68%~1.97%。结论:该方法专属性好,准确性高,可用于岩黄连总碱提取物的定性、定量分析。OBJECTIVE Qualitatively analyze the chemical constituents of total alkaloids of Corydalis saxicola Bunting,and quantitatively analyze eight alkaloids high in content.The results provide the basis for the study of pharmacodynamic material basis of total alkaloids of Corydalis saxicola Bunting,and also provide reference for its quality control.METHODS UPLC-Q-TOF-MS/MS technology was used to qualitatively analyze the total alkaloids chemical composition of Corydalis saxicola Bunting.The contents of protopine,columbamine,tetrahydropalmatine,coptisine,dehydrocavidine,palmatine,berberine and chelerythrine were determined by HPLC at the same time.RESULTS A total of 38 alkaloids were identified from the total alkaloids of Corydalis saxicola Bunting,including 13 proberberine alkaloids and 9 berberine alkaloids.In addition,the eight components of quantitative analysis had good linearity in their respective ranges(r>0.999 2);sample recovery rate 96.70%-103.8%;RSD 0.68%-1.97%.CONCLUSION The method has high specificity and accuracy,and can be used for the qualitative and quantitative analysis of total alkaloids of Corydalis saxicola Bunting.
