摘 要:目的:研究五子膏对慢传输型便秘的药理作用机制。方法:50只大鼠随机分为正常组、模型组、莫沙必利组、五子膏低剂量组,五子膏高剂量组,除正常组外,采用大黄致泻结肠法造模。正常组每天给予等体积生理盐水灌胃,其余给予相应药物干预,连续给药14d,每天2次。大鼠禁食12h,后对大鼠灌胃10%活性炭混悬液,检测小肠推进情况,采用ELISA法检测大鼠结肠NO、cGMP、PKG的表达。结果:造模后,模型组大鼠24h粪便总量、粪便含水量均明显低于正常组(P<0.05);给药后,五子膏高、低剂量组、莫沙必利组大鼠情况均有所好转;五子膏高、低剂量组和莫沙必利组大鼠粪便质地变软,24h粪便总量、粪便含水量均高于模型组(P<0.05)。模型组大鼠首次排黑便时间明显长于正常组(P<0.05),五子膏高、低剂量组、莫沙必利组首次排黑便时间明显短于模型组,各给药组大鼠炭末推进率均明显高于模型组(P<0.05)。模型组大鼠结肠NO、cGMP、PKG的表达明显高于正常组(P<0.05);五子膏高、低剂量组和莫沙必利组大鼠结肠NO、cGMP、PKG的表达均低于模型组(P<0.05)。与正常组相比,模型组大鼠血清中SP含量明显降低,VIP、SS含量明显升高;与模型组相比,莫沙必利组、五子膏高、低剂量组SP含量明显升高,VIP、SS含量显著降低。结论:给予五子膏治疗2周,大鼠慢传输型便秘现象有明显改善,肠蠕动功能增强;五子膏能显著提高血液中SP含量,降低VIP、SS含量,降低大鼠结肠组织NO、cGMP、PKG表达,进而调节大鼠胃肠道功能,促进肠道动,改善便秘。Objective:To study the pharmacological mechanism of Wuzi Gao on slow transit constipation.Method:Fifty rats were randomly divided into normal control group,model group,mosapride group,low-dose Wuzi Gao group and high-dose Wuzi Gao group.Except for the normal control group,rhubarb-induced diarrhea colon method was used for modeling.Rats in the normal group were given an equal volume of normal saline every day,and the other groups were continuously given corresponding drug interventions for 14 d,2 times a day.After fasting for 12 hours,the rats were given 10%activated carbon suspension by gavage to detect intestinal propulsion,and the expression of NO,cGMP and PKG in the colon of rats was detected by ELISA.Result:After modeling,the total fecal volume and fecal water content in 24 h in the model group were significantly lower than those in the normal group(P<0.05).After administration,the rats in high-and low-dose Wuzi Gao groups and mosapride group were all improved.The fecal texture of rats in the high-and low-dose Wuzi Gao groups and mosapride group became softer,and total fecal volume and fecal water content in 24 h were higher than those in the model group(P<0.05).The frst time of black stool excretion in the model group was significantly longer than that in the normal group(P<0.05),and it was significantly shorter in the high-and low-dose Wuzi Gao groups and mosapride group than that in the model group.The charcoal powder propulsion rate in each administration group was significantly higher than that in the model group(P<0.05).The expression of NO,cGMP and PKG in the colon of rats in the model group was significantly higher than that in the normal group(P<0.05),and it was lower in the high-and low-dose Wuzi Gao groups and mosapride group than those in model group(P<0.05).Compared with the normal control group,the content of SP in the serum of the model group was significantly decreased,and the contents of VIP and SS were significantly increased.Compared with the model group,the content of SP in the mosapride g
