摘  要:青稞鞘腐病是一种由禾生指葡孢霉(Dactylobotrys graminicola)引起的新真菌病害,目前已成为青稞主产区的重要有害生物之一.为了解青海地区青稞鞘腐病菌群体的遗传分化水平,本研究基于rDNA-ITS序列对青海青稞主产区鞘腐病菌群体的遗传多样性进行分析.结果表明:本研究分离、纯化得到89株鞘腐病菌,其群体单倍型划分为5种,单倍型H_(1)为优势单倍型,占全部菌株的95.50%,其他4个单倍型则各只有1株菌株.89株青稞鞘腐病菌群体的单倍型多样性和核苷酸多样性分别为0.110和0.01391,表明青海地区青稞鞘腐病菌群体的遗传多样性表现为低单倍型多样性与高核苷酸多样性的特征.通过邻接聚类分析得出青稞鞘腐病菌的不同单倍型未按地理来源聚类,不同地区混杂分布,表明群体遗传分化与地理来源无关.Highland barley sheath rot is a new fungal disease caused by Dactylobotrys graminicola,which has become one of the most important pests in the main producing areas of highland barley.The genetic diversity of sheath rot fungus population in the main producing areas of highland barley of Qinghai are analyzed on the basis of rDNA-ITS sequences in order to explore the genetic differentiation of highland barley sheath rot fungus in the region.The results show that there are five haplotypes of 89 strains of sheath rot pathogen population obtained by separation and purification,with H_(1) being the dominant haplotype,accounting for 95.50%of all strains,while the other four haplotypes have only one strain each.The haplotype diversity and nucleotide diversity of 89 strains are 0.110 and 0.01391 respectively,indicating that the genetic diversity characteristics of highland barley sheath rot pathogen population in Qinghai are low haplotype diversity and high nucleotide diversity.According to adjacency clustering analysis,it can be seen that different haplotypes of highland barley sheath rot pathogen are distributed in different regions without clustering in the light of geographical sources,indicating that population genetic differentiation is not related to geographical sources.
