石蝉草总木脂素含量测定方法建立及提取工艺优选

摘　　要：[目的]建立石蝉草总木脂素含量测定的方法并通过正交试验优选石蝉草总木脂素的最佳提取工艺。[方法]以五味子乙素为对照品绘制标准曲线,采用变色酸-硫酸显色体系,紫外-可见分光光度法测定石蝉草乙醇提取物中总木脂素含量;采用L_(9)(3^(4))正交试验法,以总木脂素含量为考察指标,优选石蝉草总木脂素的提取工艺。[结果]五味子乙素在4.00~24.00μg/mL线性关系良好(R^(2)=0.9961),平均回收率为97.68%,RSD为1.11%,石蝉草总木脂素优化的提取工艺为8倍量80%乙醇回流提取3次,每次2 h。[结论]该含量测定方法准确、简便、经济,适用于石蝉草总木脂素的含量测定,优选得到的提取工艺可为石蝉草总木脂素的提取提供参考。[Objective]To establish a determination method of total lignan contents from Peperomia dindygulensis and optimize the optimal ex⁃traction process of total lignans from Peperomia dindygulensis by orthogonal experiments.[Method]Schizandrin B was used as reference stand⁃ard to build the standard curve,and the content of total lignans in ethanol extract of Peperomia dindygulensis was determined by the chromo⁃tropic acid⁃H2 SO4 reaction colorimetry.Then,the optimum extraction technology was studied by the L_(9)(3^(4))orthogonal test with the total lignans of Peperomia dindygulensis as detecting index.[Result]Schizandrin B calibration curve showed a good liner relationship in 4.00-24.00μg/mL(R^(2)=0.9961),the average recovery was 97.68%,RSD was 1.11%.The optimum extraction technology of total lignans from Peperomia dindygulensis was obtained as extracting three times,two hours for each,with 8 times 80%alcohol.[Conclusion]The content determination method is accurate,simple and economical,suitable for the determination of total lignans from Peperomia dindygulensis.The optimized extraction process can provide reference for the extraction of total lignans from Peperomia dindygulensis.
