珍麻降压胶囊的提取、制备工艺优化及药效学评价

摘　　要：目的 优化珍麻降压胶囊的提取和制备工艺,并进行药效学评价。方法 研究采用L_(9)(3^(4))表正交实验设计,以干膏得率、黄芩苷、梓醇含量为评价指标,优化珍麻降压胶囊的提取工艺,以颗粒的成型率、休止角和吸湿率作为评价指标,优化珍麻降压胶囊的制备工艺,并以自发性高血压大鼠为模型,对所得珍麻降压胶囊成品的降压和镇静安神功效进行评价。结果 正交实验得出珍麻降压胶囊的最佳提取工艺为:方中牛膝、夏枯草、黄芩、生地黄、菊花、桑寄生6味药材加10倍量水,煎煮2次,每次1 h,最佳制备工艺为:稠膏与辅料比为1∶1,以80%乙醇为润湿剂,制得颗粒干燥温度为60℃。3次平行验证实验结果可见,以上提取、制备工艺均稳定可行。药效实验表明,所制得珍麻降压胶囊成品,给予临床正常服用剂量,可明显降低SHR大鼠的收缩压和舒张压,减少其自主活动次数和站立次数,该治疗效用与阳性药牛黄降压胶囊相当。结论 研究筛选出的珍麻降压胶囊提取、制备工艺稳定可行,所制得珍麻降压胶囊成品具有明确的降压及镇静安神功效。Objective To optimize the extraction and preparation technology of Zhenzhu Jiangya Capsule(珍麻降压胶囊)and evaluate its pharmacodynamics.Methods L_(9)(3^(4))orthogonal experimental design was used to optimize the extraction process of Zhenzhu Jiangya Capsule with the yield of dry paste,baicalin and catalpol content as evaluation indexes.The preparation process of Zhenzhu Jiangya Capsule was optimized with the molding rate,repose angle and moisture absorption rate as evaluation indexes.The antihypertensive and sedative effects of Zhenzhu Jiangya Capsule were evaluated in spontaneous hypertensive rats(SHR).Results Orthogonal experiments showed that the best extraction process of Zhenzhu Jiangya Capsule was as follows:Niuxi(Achyranthis Bidentatae Radix),Xiakucao(Prunellae Spica),Huangqin(Scutellariae Radix),Shengdihuang(Rehmanniae Radix),Juhua(Chrysanthemi Flos)and Sangjisheng(Taxilli Herba)added 10 times of water,decocted twice,1 h for each time.The best preparation process was as follows:the ratio of thick paste to auxiliary material was 1∶1,80% ethanol was used as wetting agent,and the drying temperature of the granules was 60℃.The results of three parallel verification experiments showed that the above extraction and preparation processes were stable and feasible.The pharmacodynamic test showed that the prepared Zhenzhu Jiangya Capsule could significantly reduce the systolic blood pressure and diastolic blood pressure of SHRs,reduce the number of autonomic activities and standing times,and the therapeutic effect was equivalent to that of the positive drug Niuhuang Jiangya Capsule(牛黄降压胶囊).Conclusion The extraction and preparation technology of Zhenzhu Jiangya Capsule screened out in this study is stable and feasible,and the finished product of Zhenzhu Jiangya Capsule has clear antihypertensive and sedative effects.
