肾衰饮对肾间质纤维化大鼠炎症与氧化应激相关基因表达谱的影响

摘  要:目的 运用网络药理学整合动物实验验证的研究思路寻找肾衰饮治疗肾间质纤维化(renal interstitial fibrosis, RIF)的可能分子机制。方法 通过中药系统药理学分析平台(TCMSP)数据库筛选肾衰饮的活性成分;NCBI-GEO数据库寻找疾病靶点,构建病药重合靶点网络及相互作用关系,然后对重合靶点进行KEGG及GO富集分析。最后采用单侧输尿管梗阻(unilateral ureteral obstruction, UUO)大鼠肾脏组织对靶点进行验证。结果 筛选出肾衰饮活性成分共计207个及7个关键重合靶点:重合靶点核转录因子-κB p65(RELA)、胞核癌基因(FOS)、基质金属蛋白酶9(MMP9)、增殖细胞核抗原(PCNA)、CYP1B1酶(CYP1B1)、2型3α-羟基类固醇脱氢酶醛酮还原酶(AKR1C3)、螺旋环螺旋结构域扩散激酶(CHUK)。动物实验表明肾衰饮能够有效减轻肾间质纤维化,肾衰饮能够显著下调小鼠肾脏组织RELA(P<0.01)、FOS(P<0.05)、MMP9(P<0.01)、PCNA(P<0.01)、CYP1B1(P<0.01)mRNA表达,上调AKR1C3(P<0.05)mRNA表达。结论 肾衰饮通过调节炎症、氧化应激等途径发挥治疗肾间质纤维化作用。Objective This study applied network-pharmacology and experiment to detect the mechanism of Shenshuai Yin(肾衰饮)treating renal interstitial fibrosis(RIF).Methods TCMSP data base was used for identifying the active ingredients of Shenshuai Yin.NCBI-GEO data base was used for finding renal failure targets and framing the network of medicine and pathology mechanism.KEGG and GO enrichment analysis was used for matching the interaction.Kidney tissue in UUO rats was taken out to verify the mechanism.Results A total of 207 active ingredients and 7 key overlapping targets,nuclear transcription factor-κB p65(RELA),FOS,matrix metalloproteinase-9(MMP9),proliferating nuclear antigen(PCNA),Cytochrome P4501B1(CYP1B1),type23 α-hydroxyl steroid dehydrogenase aldosterone reductase(AKR1C3)andconserved helix-loop-helix ubiquitous kinase(CHUK),were screened out for Shenshuai Yin.Animal experiments have shown that Shenshuai Yin can effectively alleviate renal interstitial fibrosis.Shenshuai Yin can significantly down-regulate the mRNA expressions of RELA(P<0.01),FOS(P<0.05),MMP9(P<0.01),PCNA(P<0.01)and CYP1B1(P<0.01),and up-regulate the mRNA expression of AKR1C3(P<0.05).Conclusion Shenshuai Yin rescues RIF by regulating inflammation and oxidative stress.
