摘  要:目的:探究鳖甲煎丸对肝纤维化小鼠的治疗作用及其对PPARγ表达的影响,以此阐述鳖甲煎丸治疗肝纤维化的分子机制。方法:将40只小鼠随机分为空白组、模型组、鳖甲煎丸低剂量组、鳖甲煎丸中剂量组和鳖甲煎丸高剂量组。除空白组小鼠腹腔注射橄榄油外,其余各组小鼠予四氯化碳(CCl4)腹腔注射,每周2次,连续6周。建模成功后鳖甲煎丸低、中、高剂量组小鼠给予相应剂量的鳖甲煎丸灌胃,空白组和模型组灌胃等体积生理盐水,1次/d,连续4周。实验结束后,通过Masson染色和Col-Ⅰ免疫组化观察小鼠肝脏胶原沉积,采用免疫组化检测肝脏α-SMA表达,采用蛋白质印迹法检测肝脏PPARγ表达,BSP法检测小鼠肝脏PPARγ启动子甲基化程度。结果:与空白组比较,模型组小鼠肝脏中胶原和Col-Ⅰ明显沉积(P<0.05),α-SMA表达明显升高(P<0.05),而PPARγ蛋白含量明显降低(P<0.05)。与模型组比较,鳖甲煎丸低、中、高剂量组小鼠肝脏中胶原和Col-I沉积明显减少(P<0.05),α-SMA表达明显降低(P<0.05),而PPARγ蛋白含量明显升高(P<0.05),且呈剂量依赖性(P<0.05)。BSP分析发现,模型组小鼠肝脏中PPARγ启动子甲基化位点明显多于空白组(P<0.05),鳖甲煎丸低、中、高剂量组小鼠PPARγ启动子甲基化位点明显少于模型组(P<0.05)。结论:鳖甲煎丸可通过去甲基化PPARγ启动子促进PPARγ表达,从而缓解肝纤维化。Objective:To explore the therapeutic effect of Biejiajian Pills on hepatic fibrosis in mice and its effect on PPARγexpression,so as to explain the molecular mechanism of Biejiajian Pills in the treatment of hepatic fibrosis.Methods:A total of 40 mice were randomly divided into blank group,model group,Biejiajian Pills low-dose group,Biejiajian Pills medium-dose group and Biejiajian Pills high-dose group.Except the mice in the blank group were intraperitoneally injected with olive oil,the mice in the other groups were intraperitoneally injected with carbon tetrachloride(CCl4)twice a week for 6 weeks.After successful modeling,the mice in Biejiajian Pills low,medium and high-dose groups were given the corresponding dose of Biejiajian Pills by gavage,and the blank group and the model group were given the same volume of normal saline by gavage,once a day for 4 weeks.At the end of the experiment,the collagen deposition in the liver of mice was observed by Masson staining and Col-Ⅰimmunohistochemistry,the expression ofα-SMA in the liver was detected by immunohistochemistry,the expression of PPARγin the liver was detected by Western blotting,and the methylation of PPARγpromoter in the liver of mice was detected by BSP method.Results:Compared with the blank group,the collagen and Col-Ⅰin the liver of the model group were significantly deposited(P<0.05),the expression ofα-SMA was significantly increased(P<0.05),and the expression of PPARγprotein was significantly decreased(P<0.05).Compared with the model group,the deposition of collagen and Col-Ⅰin the liver of mice in Biejiajian Pills low,medium and high-dose groups was significantly decreased(P<0.05),the expression ofα-SMA was significantly decreased(P<0.05),and the expression of PPARγprotein was significantly increased(P<0.05),and in a dose-dependent manner(P<0.05).BSP analysis showed that the methylation sites of PPARγpromoter in the liver of mice in the model group were significantly more than those in the blank group(P<0.05),and the methylation sites of
