摘  要:采用氨基酸衍生物反应试验(ADRA)对20种化学防晒剂的致敏性进行评价。利用半胱氨酸衍生物N-[2-(1-萘基)乙酰基]-L-半胱氨酸(NAC)和赖氨酸衍生物α-N-[2-(1-萘基)乙酰基]-L-赖氨酸(NAL)模拟皮肤蛋白,与化学防晒剂共同反应,通过高效液相色谱法测定20种化学防晒剂的氨基酸衍生物消耗百分比,并对皮肤致敏性进行预测。通过比较ADRA预测结果与欧盟消费者安全科学委员会(SCCS)出示的化合物评估报告的一致性,判断方法的可行性。结果表明,3-亚苄基樟脑等19种化学防晒剂预测结果为阴性,二苯酮-3预测结果为阳性。ADRA可作为动物试验体外替代方法,简便、快捷地对化合物的皮肤致敏性进行预测和筛选。The sensitization of 20 chemical sunscreens was evaluated using amino acid derivative reactivity assay(ADRA).Cysteine derivative N-[2-(1-naphthyl)acetyl]-L-cysteine(NAC)and lysine derivativeα-N-[2-(1-naphthyl)acetyl]-L-lysine(NAL)were used to mimic skin proteins,which reacted with chemical sunscreens.The amino acid derivative depletion percentage of 20 chemical sunscreens were determined by high performance liquid chromatography,and then their skin sensitization were predicted.The feasibility of ADRA method was judged by comparing the consistency of ADRA prediction results with the compound evaluation reports produced by European Scientific Committee on Consumer Safety(SCCS).It was shown that 19 chemical sunscreens such as 3-benzylidene camphor were predicted to be negative,and benzophenone-3 was predicted to be positive.ADRA could be used as an alternative in vitro method for animal experiments to predict and screen the skin sensitization of compounds simply and quickly.
