摘  要:目的使用超高效液相色谱(Ultra Performance Liquid Chromatography,UPLC)对铁皮石斛甘露糖分离色谱条件进行优化,并将色差法原理与其主要活性成分甘露糖含量相关联,探讨铁皮石斛颜色与含量的相关性,为铁皮石斛药材质量建立一种快速测定及判断的方法。方法使用UPLC测定甘露糖含量,使用CIE L^(*)a^(*)b^(*)色差法原理定量茎粉末色泽,使用R软件及SPSS进行相关性及聚类分析。结果建立了一套UPLC-TUV快速分离及测定铁皮石斛甘露糖的方法,可在8 min内使甘露糖快速出峰且完全分离,比传统高效液相色谱(High Performance Liquid Chromatography,HPLC)方法快3~10倍;铁皮石斛甘露糖含量与茎粉末明暗色度L^(*)、黄绿色度b^(*)以及总色差E^(*)ab存在极显著负相关关系(P<0.01),说明其茎粉末L^(*)值越小即偏暗黄且b^(*)值越小即偏黄棕时甘露糖含量越高(13.5%~28.9%),偏浅黄色或浅绿色时甘露糖含量越低(6.4%~12.6%)。结论采用UPLC优化铁皮石斛糖组分色谱分离条件极大地缩短甘露糖测定所需时间,可为其糖组分快速准确测定提供参考;色差原理可作为判断和评价铁皮石斛种质资源的甘露糖含量范围及品质的参考依据。Objective The separation and chromatographic conditions of mannose content in Tiepi Shihu(Dendrobium officina⁃le)were optimized by ultra high performance liquid chromatography(UPLC).The principle of color difference method was asso⁃ciated with the content of D(+)-Mannose,and the correlation between stem color and content of Tiepi Shihu(Dendrobium offi⁃cinale)was explored to establish a rapid determination for the quality of its medicinal materials.Methods UPLC was used to de⁃termine D(+)-Mannose content and the color difference was used to quantify the stem powder.R software and SPSS were used for correlation and cluster analysis.Results A set of optimal chromatographic conditions for rapid separation and determination of Tiepi Shihu(Dendrobium officinale)polysaccharide by UPLC-TUV was established.The four monosaccharides were completely separated within 8 min,which was 3~10 times faster than the traditional high performance liquid chromatography(HPLC)meth⁃od.The correlation study showed that the mannose content of Tiepi Shihu(Dendrobium officinale)was negatively correlated with the shade L^(*),yellowish green b^(*)and total color difference E^(*)ab of stem powder(P<0.01),indicating that the lower L^(*)value of Tiepi Shihu(Dendrobium officinale)powder was dark yellow,and the lower b^(*)value was yellowish brown,the higher mannose content(13.5%~28.9%).The light yellow or light green stem powder had lower mannose content(6.4%~12.6%).Conclu⁃sion The optimized chromatographic separation conditions of Tiepi Shihu(Dendrobium officinale)sugar components by UPLC greatly shortened the determination time and provided a reference for the rapid and accurate determination of sugar components.The principle of color difference can be used as a reference to judge and evaluate the mannose content range and quality of germ⁃plasm resources of Tiepi Shihu(Dendrobium officinale).
