不同煎煮方式和饮片炮制的化痰祛湿活血方药理活性对比研究

摘　　要：目的研究饮片炮制和煎煮方式对化痰祛湿活血方(Huatan Qushi Huoxue Prescription,HQHP)治疗非酒精性脂肪型肝炎(NASH)药效的影响。方法实验将SD大鼠随机分为8组:空白组、模型组、易善复组(西药阳性组)、强肝胶囊组(中药阳性组)、原方混煎组、原方单煎组、炮制混煎组、炮制单煎组,采用微板法检测血清TC、TG、GGT、ALT、AST、ALP水平,Western blot法检测肝脏ADPN、NF-κB、Akt、P-Akt、P-NF-κB水平。结果炮制前后、不同煎煮方式的HQHP均能够降低NASH大鼠血清TC、TG、GLU、ALT、AST、ALP水平(P<0.05);与原方比较,炮制后大鼠血清水平均明显降低(P<0.05),与单煎组比较,混煎组大鼠血清TC、TG、ALT水平均显著降低(P<0.05)以炮制混煎组最优。另外,与模型组比较,各治疗组大鼠均能降低NF-κB、p-NF-κB水平、升高ADPN、Akt、P-Akt水平;炮制后各实验组疗效优于原方组,混煎组优于单煎组,以炮制混煎组最优。结论饮片炮制和汤剂煎煮方法对复方药理作用有一定影响,饮片炮制和复方传统混合煎煮可以提高药理活性,为化痰祛湿活血方在临床上如何煎煮和选用炮制品提供了实验依据。Objective In order to explore the effect of processing and decoction on the efficacy of Huatan Qushi Huoxue Pre⁃scription(化痰祛湿活血方,HQHP)in the treatment of nonalcoholic steatohepatitis(NASH).Method In this experiment,SD rats were randomly divided into 8 groups:blank group,model group,Essentiale group(positive Western medicine group),Qiang⁃gan Capsule(强肝胶囊)group(positive Chinese medicine group),original formula mixed decoction group,original formula single decoction group,processed pieces mixed decoction group,processed pieces single decoction group.Serum levels of total cholester⁃ol(TC),triglyceride(TG),glutamyltransferase(GGT),alanine aminotransferase(ALT),glutamic transaminase(AST)and alka⁃line phosphatase(ALP)were detected by microplate test.The levels of adiponectin(ADPN),nuclear factor kappa-B(NF-κB),protein kinase B(Akt),phosphorylated Akt protein(P-Akt),phosphorylation nuclear factor-κB(P-NF-κB)in liver were detected by Western Blot method.Result The levels of TC,TG,GLU,ALT,AST and ALP in serum of NASH rats were de⁃creased by HQHP with different decoction methods before and after processing(P<0.05).Compared with those of the original formula groups,the serum levels of the processed pieces groups were significantly decreased(P<0.05).Compared with those of the single decoction groups,the serum levels of TC,TG and ALT in the mixed decoction groups were significantly decreased(P<0.05).In addition,compared with the model group,the rats in each treatment group can reduce the levels of NF-κB and p-NF-κB,and increase the levels of ADPN,Akt and P-Akt.The processed pieces groups were better than the original formula groups,and the mixed decoction groups were than the single decoction groups and the processed pieces mixed decoction group was the best.Conclusion The preparation of decoction pieces and decoction methods have a certain influence on the pharmacological effects of traditional Chinese medicine compounds.The preparation of decoction pieces and the traditional mixed decoct
