石萆汤对环磷酰胺干预后大鼠精子DNA碎片率及性激素的影响

摘　　要：目的研究石萆汤对环磷酰胺干预后大鼠精子的DNA碎片率及性激素的影响。方法随机选取8只大鼠为正常组,其余32只大鼠为造模组,造模组连续14 d腹腔注射环磷酰胺(20 mg·kg^(-1)·d^(-1))制备大鼠少弱精子症模型,然后将造模组随机分为模型组、石萆汤低、中、高剂量(6.5625 g/kg、13.125 g/kg、26.25 g/kg)组,每组8只。正常组和模型组给予生理盐水灌胃,石萆汤低、中、高剂量组分别给予相对应的中药剂量灌胃,中药干预35 d后,采用流式细胞仪检测大鼠精子DNA碎片率(DFI);酶联免疫吸附测定法(ELISA法)检测血清中卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(T)、泌乳素(PRL)、雌激素(E_(2))的水平。结果与正常组比,模型组大鼠精子DFI显著增高(P<0.01);与模型组相比,治疗后石萆汤低、中、高剂量组的大鼠精子DFI显著下降(P<0.01);血清性激素水平检测结果显示,模型组T、FSH和LH含量显著降低(P<0.01);E2和PRL含量显著升高(P<0.01);石萆汤低、中、高剂量组均能改善大鼠血清性激素水平,以中剂量最为明显,尤其是提高T的水平(P<0.01)。结论石萆汤能平衡环磷酰胺干预后大鼠的性激素水平,降低大鼠的精子DFI,为临床研究提供实验依据和基础。Objective To study the effects of Shibi decoction on sperm DNA fragmentation rate and sex hormone of rats after cyclophosphamide intervention.Methods Eight rats were randomly selected as the normal group and the other 32 rats as the model group.The rat models with oligoasthenospermia were established by intraperitoneal injection of cyclophosphamide(20 mg·kg^(-1)·d^(-1))for 14 days.Then in the model rats group were randomly divided into the model group,low,medium and high dose groups of ShiBi decoction(6.5625 g/kg,13.125 g/kg,26.25 g/kg),with 8 rats in each group.The rats in the normal group and the model group were treated by gavage with normal saline,and the rats in the low,medium and high dose groups of ShiBi decoction with corresponding traditional Chinese medicine,respectively.After 35 days of traditional Chinese medicine intervention,the sperm DNA fragment rate(DFI)of rats was detected by flow cytometry,serum levels of follicle stimulating hormone(FSH),luteinizing hormone(LH),testosterone(T),prolactin(PRL)and estrogen(E_(2))were detected by enzyme⁃linked immunosorbent assay(ELISA).Results Compared with that in the normal group,the rate of sperm DNA fragments in the model group was significantly increased(P<0.01);Compared with that in the model group,the sperm DNA fragment rates of rats in the low,medium and high dose group of ShiBi decoction after treatment were all decreased significantly(P<0.01);Analysis of serum sex hormones showed that the levels of T,FSH and LH in the model group were decreased significantly(P<0.01),but the levels of E2 and PRL were increased significantly(P<0.01);After ShiBi decoction intervention,the levels of serum sex hormones in rats,especially the level of T were all elevated in the low,medium and high dose groups(P<0.01).Conclusion After cyclophosphamide intervention,Shibi decoction can balance sex hormone level and reduce the sperm DFI of rats.Our data provide experimental basis for clinical application research.
