摘  要:为评估五倍子颗粒(Galla Chinensis granules,GCG)止泻、抗炎和镇痛的药效作用,通过番泻叶诱导小鼠腹泻模型,二甲苯诱导耳肿胀及角叉菜胶诱导足肿胀急性炎症模型,以及小鼠疼痛模型(热板法和扭体法)的构建,观察GCG对小鼠腹泻、胃肠运动、炎症和痛觉反应的影响,并通过病理组织学及血清炎性细胞因子检测,进一步探究GCG的药效作用。结果表明,与模型组比较,GCG治疗组(50 mg/kg BW、100 mg/kg BW和200 mg/kg BW)对小鼠腹泻具有显著抑制作用(P<0.05),可极显著降低小鼠胃肠蠕动(P<0.01),且对小鼠耳肿胀及足肿胀具有显著抑制作用(P<0.05),极显著抑制醋酸引起的腹部收缩(P<0.01),其中,GCG治疗组(200 mg/kg BW和100 mg/kg BW)分别在给药后0.5 h和1.0 h开始延长小鼠热板试验中热刺激引起的疼痛反应时间(P<0.05),而GCG治疗组(50 mg/kg BW)只在4.0 h延长小鼠疼痛反应时间(P<0.05)。其次,GCG治疗组均可不同程度改善角叉菜胶引起的足病理变化,且GCG治疗组血清炎症因子TNF-α、PGE2及COX-2水平显著低于模型组(P<0.05)。综上,GCG止泻、抗炎及镇痛作用较为显著,对小鼠胃肠蠕动、腹泻及炎症反应具有抑制作用,且能够减轻小鼠疼痛反应,可为其临床应用提供依据。The objective of this study was to evaluate the antidiarrheal,anti-inflammatory,and analgesic effects of Galla Chinensis granules(GCG)in mice.Mice diarrhea models induced by senna leaf,inflammatory model of xyleneinduced ear swelling and carrageenan-induced foot swelling,and mice pain models including hot plate test and acetic acid-writhing test,were established,and the effects of GCG on diarrhea,gastrointestinal motility,inflammation and pain response in mice were observed,and the pharmacological effects of GCG were further elucidated by histopathology and serum inflammatory cytokine indexes.Compared with the control,mice treated with GCG at doses of 50 mg/kg BW,100 mg/kg BW and 200 mg/kg BW showed significantly suppressed diarrhea(P<0.05),inhibited gastrointestinal motility(P<0.01),ear and paw swelling(P<0.05),and abdominal contraction-induced acetic acid(P<0.01).The duration of pain response induced by thermal stimulation in the hot plate test was prolonged in mice starting 0.5 h and 1.0 h after treatment with GCG(200 mg/kg BW and 100 mg/kg BW)(P<0.05).In contrast,mice treated with GCG(50 mg/kg BW)had prolonged pain response time only at 4.0 h(P<0.05).GCG could ameliorate carrageenan-induced pathological changes in the paw,and serum inflammatory cytokines(TNF-α,PGE2 and COX-2)levels were lower in the GCG-treated mice than that in the model group(P<0.05).In conclusion,GCG can inhibit the gastrointestinal peristalsis,diarrhea,and inflammatory response in mice,and reduce the pain response in mice,thus GCG may have a beneficial efficacy in the antidiarrheal,anti-inflammatory,and analgesic effects of GCG are significant,which can provide experimental basis for clinical application.
