摘 要:流感病毒的复制会同时受到宿主细胞因子的协助与拮抗。宿主细胞因子在应对流感病毒所表现出来“顺从与反抗”的两面性是未来针对宿主而非病毒本身的抗病毒药物设计的理论基础。这一概念也被称为宿主导向疗法(host-directed therapeutics, HDTs)。利用基因筛查技术已经筛选出能够调节流感病毒感染的宿主因子,这些因子可作为宿主导向抗病毒策略的候选靶点。然而,尽管在了解宿主靶点方面取得了进展,但是目前还没有可用于临床中针对流感病毒的宿主导向干预措施。就上述存在的问题,本篇综述将系统阐述基于基因敲除/敲低以及过表达筛选中发现的一些宿主因子的生物学特性,并且讨论分析这些因子用于宿主导向的抗流感策略中的潜在价值。During replication,influenza viruses are simultaneously supported and antagonized by various factors derived from host cells.These supportive and antagonistic forces of host factors are a major topic in influenza infection studies,to aid in the future design and development of novel drugs.This concept is termed host-directed therapeutics(HDTs).Through genetic screening,multiple host factors that modulate influenza infections have been identified,and may serve as candidate targets for host-directed antiviral strategies.Despite the importance of host-directed antiviral strategies,few host-directed interventions are currently available for clinical applications against influenza viruses.In light of the challenges mentioned above,this review describes the biological characterization of host factors identified via knockout/knockdown and over-expression screens,and discusses the potential use of host factors in clinical applications against influenza.
