摘  要:目的:观察洪格日-12超微粉对大鼠发热模型、炎症模型及小鼠疼痛模型的解热、抗炎、镇痛作用。方法:采用干酵母诱导建立发热模型,观察洪格日-12超微粉解热作用;光照甩尾试验和冰醋酸扭体试验,观察洪格日-12超微粉镇痛作用;二甲苯致耳廓肿胀试验和角叉菜胶足趾肿胀试验,观察超微粉抗炎作用;用ELISA法检测角叉菜胶足趾肿胀试验血清中白细胞介素6(IL-6)、IL-10、肿瘤坏死因子α(TNF-α)和前列腺素E2(PGE_(2))含量及检测组织病理形态;采用免疫组织化学法测定足组织中细胞外调节蛋白激酶(ERK1/2)、环氧合酶-2(COX-2)免疫阳性细胞表达和免疫印迹法检测细胞外调节蛋白激酶(ERK1/2)及磷酸化细胞外调节蛋白激酶(p-ERK1/2)在足组织中的表达。结果:与模型对照组比较,洪格日-12预防给药7 d造模后5 h体温明显降低(P<0.05);与模型对照组比较洪格日-12超微粉组甩尾时间明显延长,扭体次数、耳廓肿胀率明显降低(P<0.05);洪格日-12超微粉1 g/kg组镇痛率达到了51.67%(P<0.05);与模型对照组比较,洪格日-12超微粉0.15、0.3、0.6 g/kg组各时间点足肿胀率明显降低(P<0.05)。与正常对照组比较,模型对照组大鼠血清IL-6、IL-10及TNF-α、PGE_(2)含量明显升高,ERK1/2及COX-2阳性细胞分布数量及密度明显增加(P<0.05或P<0.01);与模型对照组比较,给药组大鼠血清IL-6、IL-10及TNF-α、PGE_(2)含量明显降低(P<0.05);模型对照组足组织有明显炎性细胞,各给药组炎性细胞均减少,ERK1/2及COX-2阳性表达明显减少(P<0.05或P<0.01)。免疫印迹法检测结果表明,随着超微粉剂量增加,ERK1/2磷酸化表达明显下调(P<0.05)。结论:蒙药洪格日-12超微粉具有明显的抗炎、镇痛作用,其抗炎机制可能与抑制炎性因子IL-6、IL-10、TNF-α、PGE_(2)的表达及ERK1/2的磷酸化表达和下调ERK1/2、COX-2蛋白的表达有关。Objective:The antipyretic,anti-inflammatory,and analgesic effects of honggeri-12 ultra-micro powder on the rat fever model,inflammation model,and mice pain model were observed.Methods:The fever model was induced by dry yeast to observe the antipyretic effect of honggeri-12 ultra-micro powder.The analgesic effect of honggeri-12 ultra-micro powder was observed in the light tail flicking test and glacial acetic acid twisting test.Xylene-induced auricle swelling test and carrageenan toe swelling test were performed to observe the anti-inflammatory effect of the ultra-micro powder.The content of interleukin 6(IL-6),interleukin 10(IL-10),tumor necrosis factor a(TNF-α),and prostaglandin E2(PGE_2) in the serum of carrageenan toe swelling experiment was detected by enzyme-linked immunosorbent assay(ELISA),and histopathological morphology was observed.Immunohistochemistry was used to detect extracellular regulatory protein kinase(ERK1/2) and cyclooxygenase-2(COX-2) immunopositive cells.Western blotting was used to detect the expressions of ERK1/2 and phosphorylated ERK1/2(p-ERK1/2) in foot tissues.Results:Compared with the model control group,the body temperature was reduced 5 h after 7 days of prophylactic administration of Hongeri-12(P<0.05).Compared with that in the model control group,the tail flicking time in honggeri-12 ultra-micro powder group was significantly prolonged,and the number of twisting times and ear swelling rate were significantly reduced(P<0.05).In honggeri-12 ultra-micro powder 1 g/kg group,the analgesic rate reached 51.67%(P<0.05).Compared with the model control group,the foot swelling rates of different time points in Hongeri-12 ultra-micro powder groups(0.15,0.3,and 0.6 g/kg) were significantly decreased(P<0.05).Compared with the control group,the contents of IL-6,IL-10,TNF-α,and PGE_(2) were significantly increased in the model control group.Compared with the model group,the contents of IL-6,IL-10,TNF-α,and PGE_(2) were significantly decreased in the administration groups(P<0.05).Obvious inflam
