摘 要:目的:探讨牵牛子水煎液及化学拆分组分对大鼠上焦水饮内停模型药效学研究,寻找牵牛子对上焦水饮内停模型作用的药效物质基础。方法:采用复合造模因素(大鼠腹部皮下注射异丙肾上腺素+气管插管+寒冷刺激)建立上焦水饮内停模型。模型建立后开始给予牵牛子水煎液0.63 g/kg、多糖组分0.37 g/kg、酚酸组分0.13 g/kg、树脂苷组分0.10 g/kg、脂肪油组分0.03 g/kg,灌胃给药连续14 d。试验结束后检测大鼠血清白蛋白(ALB)、肌酸激酶(CK)含量,计算肺干湿比,心、肺脏指数,测定肺泡灌洗液中蛋白含量,观察心、肺组织病理切片,综合评价牵牛子水煎液及不同组分对上焦水饮内停模型大鼠利水功效。结果:与正常对照组比较,模型对照组ALB含量显著降低(P<0.01),肺泡灌洗液中蛋白浓度、肺干湿比、CK含量以及心脏、肺脏指数显著升高(P<0.01),心、肺组织受损较严重,上焦水饮内停模型建立成功;与模型对照组比较,牵牛子水煎液0.63 g/kg组显著升高ALB含量(P<0.01),降低心脏指数、肺脏指数、肺干湿比、肺泡灌洗液中蛋白含量(P<0.05或P<0.01);树脂苷0.10 g/kg组可降低CK含量、心脏指数、肺脏指数、肺干湿比、肺泡灌洗液中蛋白含量(P<0.05或P<0.01);多糖0.37 g/kg组明显升高ALB含量(P<0.05)。牵牛子水煎液0.63 g/kg组、树脂苷0.10 g/kg组对上焦水饮内停模型大鼠心、肺病理改变有明显改善作用。结论:牵牛子水煎液可改善上焦水饮内停模型大鼠的心肺损伤。树脂苷组分治疗作用明显,为牵牛子对上焦水饮内停模型大鼠具有治疗作用的药效物质基础。Objective:To explore the pharmacodynamical effects of the water decoction and chemically fractionated components of PHARBITIDIS SEMEN on a rat model of Shangjiao Shuiyinneiting(上焦水饮内停)and identify the pharmacodynamic material basis.Methods:The rat model of Shangjiao Shuiyinneiting was established using multiple factors(subcutaneous injection of isoproterenol,tracheal intubation,and cold stimulation).After model establishment,the rats were orally administered with PHARBITIDIS SEMEN water decoction at a dose of 0.63 g/kg,polysaccharide fraction at 0.37 g/kg,phenolic acid fraction at 0.13 g/kg,resin glycoside fraction at 0.10 g/kg,and fatty oil fraction at 0.03 g/kg for 14 consecutive days.At the end of the experiment,the levels of serum albumin(ALB),creatine kinase(CK),lung wet-to-dry ratio(W/D),protein content in bronchoalveolar lavage fluid,as well as heart and lung indices,and histopathological sections of the heart and lungs were measured to comprehensively evaluate the Lishui(利水)effect of PHARBITIDIS SEMEN water decoction and its different components on the rat model of Shangjiao Shuiyinneiting.Results:Compared with the normal control group,the model control group showed a significant decrease in ALB content(P<0.01)and a significant increase in protein concentration in bronchoalveolar lavage fluid,lung W/D,CK content,as well as heart and lung indices(P<0.01),indicating severe damage to the heart and lungs,and successful establishment of the rat model of Shangjiao Shuiyinneiting.Compared with the model control group,the group treated with PHARBITIDIS SEMEN water decoction at a dose of 0.63 g/kg showed a significant increase in ALB content(P<0.01)and a decrease in heart and lung indices,lung W/D,and protein content in bronchoalveolar lavage fluid(P<0.05 or P<0.01).The group treated with the resin glycoside fraction at 0.10 g/kg showed a decrease in CK content,heart and lung indices,lung W/D,and protein content in bronchoalveolar lavage fluid(P<0.05 or P<0.01).The group treated with the polysaccha
