摘  要:目的:探讨天南星胆汁制前后的寒热药性变化,阐明相反为制的天南星由温燥转为寒凉的科学性。方法:建立大鼠寒热证模型,观察大鼠进食量、饮水量、肛温、体质量的变化和结肠的病理变化,检测大鼠血清中的多巴胺(DA)、环磷酸腺苷(cAMP)、甲状腺激素(T4)、去甲肾上腺素(NE)、5-羟色胺(5-HT)、促甲状腺素释放激素(TRH)、促甲状腺激素(TSH)、17-羟皮质类固醇(17-OHCS)的含量,大鼠肝脏中丙酮酸(PA)、ATP酶活力;免疫组化观察大鼠海马、下丘脑、背根神经节中热敏感蛋白(TRPV1)和寒敏感蛋白(TRPM8)的表达,研究胆南星炮制前后对寒热模型大鼠的影响。结果:天南星2.8 g/kg可升高寒证大鼠血清中NE、T4、DA、TSH、TRH、cAMP、17-OHCS含量和肝脏中丙酮酸(PA)、Na^(+)-K^(+)/ATP酶、Ca^(2+)-Mg^(2+)/ATP酶含量,降低5-HT含量,改善寒证大鼠结肠损伤,下调海马、下丘脑、背根神经节中TRPM8的表达(P<0.05或P<0.01);胆南星和胆汁可以降低热证大鼠血清中NE、T4、DA、TSH、TRH、cAMP、17-OHCS含量和肝脏中PA、Na^(+)-K^(+)/ATP酶、Ca^(2+)-Mg^(2+)/ATP酶含量(P<0.05或P<0.01);升高5-HT含量,改善热证大鼠结肠损伤,下调海马、下丘脑、背根神经节中TRPV1的表达(P<0.05或P<0.01)。结论:天南星炮制成胆南星后药性由温热转为寒凉,胆汁药性属寒凉。Objective:To explore the changes of Han-Re(寒热)medicinal properties before and after preparation of Arisaematis Rhizoma with bile,and to explain the scientific nature of Arisaematis Rhizoma from Wenzao(温燥)to Hanliang(寒凉)in the opposite way.Methods:A rat model of Han-Re syndrome was established.Serum levels of dopamine(DA),adenosine phosphate(cAMP),thyroid hormone(T4),norepinephrine(NE),5-hydroxytryptamine(5-HT),thyrotropin-releasing hormone(TRH),thyroid-stimulating hormone(TSH)and 17-hydroxycorticosteroid(17-OHCS)were detected,and thepyruvate(PA)content and ATPase activity were detected in rat liver.The protein expressions of transient receptor potential vanilloid 1(TRPV1)and transient receptor potential cation channel subfamily M member 8(TRPM8)in rat hippocampus,thalamus and dorsal root ganglion were determined.At the same time,the changes in food intake,water intake,rectal temperature and body weight and the pathological changes of colon were detected to study the effect of Arisaema Cum Bile in Han-Re syndrome rats before and after processing.Results:On Han(寒)syndrome rat,Arisaematis Rhizoma could increase the serum levels of NE,T4,DA,TSH,TRH,cAMP,17-OHCS,and the contents of PA and activities of Na^(+)-K^(+)/ATPase and Ca^(2+)-Mg^(2+)/ATPase in liver.It also reduced 5-HT content,alleviated the colon injury,and down-regulated the expression of TRPM8 in the hippocampus,hypothalamus,and dorsal root ganglion of rats with Han syndrome.In Re(热)syndrome rats,Arisaema Cum Bile and bile reduced the serum levels of NE,T4,DA,TSH,TRH,cAMP,17-OHCS,and contents of PAand activities of Na^(+)-K^(+)/ATPase and Ca^(2+)-Mg^(2+)/ATPase in liver.It also increased the 5-HT content,alleviated the colon injury,and down-regulated the expression of TRPV1 in the hippocampus,hypothalamus,and dorsal root ganglion of rats with Re syndrome.Conclusion:After Arisaematis Rhizoma was processed into the bile product,its medicinal property was changed from Wenre(温热)to Hanliang.The medicinal property of bile was Hanliang.
