摘 要:为了解地黄低聚糖对6种益生菌体外增殖的影响,试验将嗜酸乳杆菌、发酵乳杆菌、动物双歧杆菌、长双歧杆菌婴儿亚种、短双歧杆菌和罗伊氏乳杆菌6种益生菌分别用含0.5%、1%、2%、4%地黄低聚糖(作为碳源)的培养基培养,以不含葡萄糖的MRS肉汤培养基为阴性对照,以含2%葡萄糖的MRS肉汤培养基为阳性对照,测定地黄低聚糖培养6种益生菌的生长曲线、pH值,根据生长情况选取其中一个发酵效果最好的地黄低聚糖浓度检测发酵液中短链脂肪酸(乙酸、丙酸、异丁酸、丁酸、戊酸和异戊酸)生成情况与糖类成分(果糖、蔗糖、葡萄糖、棉籽糖、水苏糖、毛蕊花糖)的变化。结果表明:6种益生菌均可利用不同浓度地黄低聚糖进行增殖,与2%葡萄糖培养基相比,各菌种可优先利用地黄低聚糖进行增殖,其中2%、4%地黄低聚糖增殖效果优于其他浓度,6种益生菌对短双歧杆菌和罗伊氏乳杆菌增殖效果较好,优于其他益生菌;以经济适用原则选取2%地黄低聚糖进行后续试验,2%地黄低聚糖发酵液中短链脂肪酸生成均以乙酸为主,但糖类成分的含量存在差异,短双歧杆菌和罗伊氏乳杆菌发酵液中主要为葡萄糖,而其他4种发酵液中除葡萄糖外还含有较多的棉籽糖、蔗糖。说明地黄低聚糖可促进6种益生菌增殖,但增殖效果不同,其中短双歧杆菌和罗伊氏乳杆菌增殖效果最佳。In order to understand the effects of Rehmannia glutinosa oligosaccharides on the proliferation of six probiotics in vitro,in this experiment,six probiotics namely Lactobacillus acidophilus,Limosilactobacillus fermentum,Bifidobacterium animalis,Bifidobacterium longum subsp.infantis,Bifidobacterium breve,and Limosilactobacillus reuteri,were cultured respectively in medium containing 0.5%,1%,2%and 4%Rehmannia glutinosa oligosaccharides as carbon sources;glucose-free MRS broth medium was used as the negative control and MRS broth medium containing 2%glucose was used as the positive control;the growth curve and pH value of Rehmannia glutinosa oligosaccharides were determined.One concentration of Rehmannia glutinosa oligosaccharides with the best fermentation effect was selected for the determination of the production of short-chain fatty acids(acetic acid,propionic acid,isobutyric acid,butyric acid,valeric acid and isovaleric acid)and the changes of carbohydrate composition(fructose,sucrose,glucose,raffinose,stachyose,and verbascose)in the fermentation broth.The results showed that when cultured with different concentrations of Rehmannia glutinosa oligosaccharides,the six probiotics could be proliferated by Rehmannia glutinosa oligosaccharides.Compared with 2%glucose culture medium,each type of the bacteria could preferentially use Rehmannia glutinosa oligosaccharides for proliferation,and the proliferation effect of 2%and 4%Rehmannia glutinosa oligosaccharides was better than that of other concentrations.Among the six probiotics,the proliferative effect on Bifidobacterium breve and Limosilactobacllus reuteri was better than other probiotics.Based on the principle of economic applicability,2%Rehmannia glutinosa oligosaccharides were selected for follow-up tests.The formation of short-chain fatty acids in the fermentation broth of 2%Rehmannia glutinosa oligosaccharides was mainly acetic acid,but there were differences in the contents of carbohydrates.The fermentation broth of Bifidobacterium breve and Limosilactobacil
