摘 要:以单细胞RNA测序(single cell RNA sequencing,scRNA-seq)和单细胞核RNA测序(single-nucleus RNA sequencing,snRNA-seq)进行单细胞/单细胞核转录组学分析,为组织在稳态维持或动态变化过程(如疾病的发展、分化和发育)中提供了一个全面的组织细胞统计可视图。与传统批量分析相比,单细胞/单细胞核为中心的转录组分析更准确地揭示组织内细胞的异质性和细胞内基因调控网络。该文总结了应用sc/snRNA-seq技术分析心脏细胞转录谱的新发现,以及心血管发育的重要新分子机制。Single-cell transcriptomics analysis with single cell and single nucleus RNA sequencing(sc/snRNA-seq)provides a global and unbiased view of tissues cellular demographics both at steady-state and in dynamic processes such as development,differentiation,and progression of diseases.Single cel/nucleus transcriptome analysis reveals the cell heterogeneity more accu-rately than the traditional batch analysis,as well as the regulation network in cells.In this review,we summarize the recent find-ings in the cardiac cellular atlas from various sc/snRNA-seq platforms and describe the latest progress in molecular mechanisms of cardiovascular development and cardiac regeneration.
