胺鲜脂对箱根草组培快繁的影响

摘  要:为解决箱根草组培苗增殖和生根效率低难以实现规模化生产的问题,将胺鲜脂与6-BA、NAA和IBA复配使用,进行了箱根草芽增殖和生根培养的试验研究。结果表明:当添加4.0 mg·L^(-1)胺鲜脂时,芽增殖可达11.3倍,株高达到2.5 cm以上的苗占比69.0%,表现效果最佳;添加1.0 mg·L^(-1)胺鲜脂时,促进生根效果最佳,生根率达到100%,培养25 d时具有10条及以上根数的苗占比达73.3%。将6-BA、NAA和IBA与胺鲜脂复配使用后,有效促进了其效能发挥,使箱根草不定芽增殖倍数由6.7增加到11.3,增殖培养后符合生根标准的苗增加134.2%,生根率提高达45.6%,使箱根草的增殖、生长和生根效果更好,大幅提高了生产效率和苗的质量,使生产成本降低66.5%。上述研究成功建立箱根草组培快繁技术体系,实现了低成本生产优质种苗。In order to solve the problem of low proliferation and rooting efficiency in tissue culture seedlings of Hakonechloa macra'Aureola',which is difficult to achieve large-scale production,a combination of 6-BA,NAA,and IBA with DA-6 was used to conduct experimental studies on bud proliferation and rooting culture.The results showed that when 4.0 mg·L^(-1)DA-6 was added,the proliferation of buds could reach 11.3 times,and 69.0%of seedlings with a height of over 2.5 cm accounted for the best performance;When 1.0 mg·L^(-1)DA-6 was added,the best effect was to promote rooting,with a rooting rate of 100%.After 25 days of cultivation,the proportion of seedlings with 10 or more roots reached 73.3%.After the combination of 6-BA,NAA,and IBA with DA-6,their efficiency was effectively promoted,resulting in an increase in the proliferation rate of adventitious buds from 6.7 to 11.3.After proliferation and cultivation,the number of seedlings that meet the rooting standards increased by 134.2%,and the rooting rate increased by 45.6%.This improved the proliferation,growth,and rooting effects of Hakonechloa macra'Aureola',significantly improving production efficiency and seedling quality,and reducing production costs by 66.5%.This study successfully establishes a tissue culture and rapid propagation technology system for Hakonechloa macra'Aureola',achieving low-cost production of high-quality seedlings.
