摘  要:目的研究高原多脏器功能障碍综合征(H-MODS)蕨麻猪模型内皮型一氧化氮合成酶(eNOS)和内皮素1(ET-1)在其炎性反应发展过程中的变化特点。方法选用低、中、高3个剂量(B、C、D组)内毒素(LPS)诱导高原土生蕨麻猪成功建立H-MODS模型,0.9%氯化钠溶液为对照组(A组)。检测不同时点血清eNOS及ET-1表达水平。结果各组蕨麻猪泵入LPS后eNOS和ET-1表达量均急剧升高,并在3 h时升至最高,之后快速下降,至24 h基本恢复到正常值,对照组各时间点表达量无显著性变化。eNOS表达量B、C、D组在3~12 h与0 h和A组相比明显升高(P<0.01);24~72 h B、C、D组eNOS表达量基本恢复到正常水平,与0 h和A组相比无明显差异。ET-1表达量B、C、D组在3~6 h与0 h和A组相比显著升高(P<0.01);B、C组ET-1表达量在12 h与0 h和A组相比明显升高(P<0.05);D组ET-1表达量在12 h与A组相比明显升高(P<0.05);24~72 h B、C、D组ET-1表达量基本恢复到正常值,与0 h和A组相比无明显差异。结论高原多脏器功能障碍综合征(MODS)形成时机体产生严重的炎性反应,eNOS及ET-1表达量迅速升高,但在免疫调节作用下其表达量迅速出现下调达到平衡状态。Objective To study the changes of endothelial nitric oxide synthase(eNOS)and endothelin-1(ET-1)in the development of inflammation in Juema pig model of high altitude multiple organ dysfunction syndrome(H-MODS).Method H-MODS model was successfully established in plateau native Juema pigs induced by endotoxin(LPS)at three doses of low,medium and high(groups B,C and D).Normal saline was used as the control group(group A).The expression levels of serum eNOS and ET-1 were detected at different time points.Result The expression levels of eNOS and ET-1 increased sharply after LPS was pumped into Juema pigs in each group,and rose to the highest at 3 h,then decreased rapidly,and basically returned to the normal value at 24 h.There was no significant change in the expression levels at each time point in the control group.The expression of eNOS in group B,C and D was significantly higher than that in group A and group 0 h at 3-12 h(P<0.01);at 24-72 h,the expression of eNOS in groups B,C and D basically returned to the normal level,and there was no significant difference compared with group 0 h and group A.The expression of ET-1 in group B,C and D was significantly higher than that in group A and group 0 h at 3-6 h(P<0.01);the expression of ET-1 in group B and C was significantly higher than that in group A and group 0 h at 12 h(P<0.05);the expression of ET-1 in group D was significantly higher than that in group A at 12 h(P<0.05);the expression of ET-1 in groups B,C and D basically returned to normal at 24-72 hours,and there was no significant difference compared with group 0 h and group A.Conclusion During the formation of plateau mods,the body produces serious inflammatory reaction,and the expression of eNOS and ET-1 increases rapidly,but the expression decreases rapidly under the effect of immune regulation,and its changes reach a balance state after a sharp rise and fall in a short time.
