摘  要:为探索果桑种质之间的亲缘关系,选取适宜海南地区生长的34份果桑资源,通过可靠的SRAP分子标记技术,进行果桑种质的遗传多样性分析研究。结果表明:从100对SRAP引物组合中筛选获得16对扩增多态性高的引物组合,共扩增清晰的条带108条,其中多态性条带93条,平均多态性比率为84.96%,平均每对引物组合扩增6.75条。平均观测等位基因数为1.8704,平均有效等位基因数为1.5200,平均Nei′s基因多样性指数为0.3022,平均Shannon′s信息指数为0.4510。遗传相似系数为0.49~0.95,表明34份果桑种质之间遗传多样性比较高。根据UPGMA聚类树状图,在遗传相似系数为0.32处,供试34份果桑种质可归属为五大类群,其中第Ⅰ类群27份,占79.41%,包括大部分果桑供试材料;第Ⅱ类群3份种质,占8.82%,分别为嘉陵30号、云桑2号、本地桑HNQZ01(采自海南琼中);第Ⅲ类群和第Ⅳ类群各1份种质,分别为红宝石和长果桑,均占到2.94%;第Ⅴ类群包括2份种质,分别为香金葚、长相思,占5.88%。本研究结果可为果桑种质资源的鉴定、评价及优良品种选育提供科学依据。In order to explore the genetic relationships among fruit mulberry germplasms,34 fruit mulberry germplasm resources suitable for growth in Hainan were selected to analyze the genetic diversity of fruit mulberry germplasm by using reliable SRAP molecular marker technology.The results showed that 16 pairs of highly polymorphic primers were selected from 100 pairs of SRAP primer combinations.A total of 108 clear bands were amplified,including 93 polymorphic bands,with an average polymorphism rate of 84.96%,and an average of 6.75 bands were amplified per pair of primer combinations.The average observed allele number was 1.8704,the average effective number of alleles was 1.5200,the average Nei′s genetic diversity index was 0.3022,the average Shannon′s information index was 0.4510.The genetic similarity coefficient was 0.49-0.95,indicating that the genetic diversity of 34 fruit mulberry germplasm was relatively high.According to UPGMA clustering tree map,34 fruit mulberry germplasm samples could be classified into five groups at the genetic similarity coefficient of 0.32,among which,27 samples were from GroupⅠ,accounting for 79.41%,including most of the fruit mulberry materials.Three germplasm of GroupⅡ,which were Jialing No.30,Yunsang No.2 and local mulberry HNQZ01(collected from Qiongzhong,Hainan),accounting for 8.82%.There was one germplasm from groupⅢand one germplasm from groupⅣ,which were Hongbaoshi and Changguosang,accounting for 2.94%.The groupⅤincluded 2 germplasm,namely Xiangjinshen and Changxiangsi,all accounting for 5.88%.The results can provide scientific basis for the identification,evaluation and breeding of fruit mulberry germplasm resources.
