摘 要:采用响应面法优化微波提取怀牛膝多糖的条件。在单因素试验的基础上,选取微波提取功率、提取时间、料液比和提取次数为影响因子,应用Box-behnken中心组合进行四因素三水平的试验设计,以怀牛膝多糖得率作为响应值,进行响应面分析(RSM)。结果表明,微波法提取多糖的最佳工艺参数为提取功率800 W,提取时间2.5 min,料液比1∶30(g∶mL),提取次数2次,模型预测值为29.38%,实际测得多糖得率为29.40%,与模型预测值的相对误差为0.07%,模型预测值为与模型预测值基本相符。微波辅助法适用于提取怀牛膝多糖。Response surface analysis methodology(RSM)was used for optimizing microwave-assisted extraction of Achyranthes bidentata Bl polysaccharides(ABP).Based on single-factor experiments,four independent variables,namely microwave power,extraction time,liquit-to-solid ratio and extraction times were selected as casual factors.The experiments were arranged accord ing to Box-behnken central composite experiment design.Response surface analysis was applied to determine the effect of central composite experiment design.Response surface analysis was applied to determine the effect of casual factors on the yield of Achyranthes bidentata Bl polysaccharides.The results showed that the optimum microwave-assisted extraction condition for Achyranthes bidentata Bl polysaccharides was as follows:microwave power 800 W,extraction time was 2.5 min,liquid to material ratio 1∶30(g∶mL),and number of extractions was 2.The predicted best extraction yield was 29.38%,while actual extraction yield was 29.40%,with relative error 0.07%,which was consistent with the predicted results.These results indicated that microwave-assisted method was suitable for extraction polysaccharides from the Achyranthes bidentata Blume.
