摘 要:目的:探讨表皮生长因子(EGF)对老年大鼠自发运动活性及黑质-纹状体多巴胺体系(NSDAs)的影响。方法:经皮给予健康24月龄大鼠EGF(0.15 mg/mL)7 d,通过旷场试验检测大鼠的自发运动活性;通过实时定量PCR和免疫印迹检测NSDAs中酪氨酸羟化酶(TH)和多巴胺转运体(DAT)的表达;通过液相色谱-串联质谱分析NSDAs中多巴胺(DA)的含量变化。结果:老年组大鼠自发运动活性、NSDAs中TH和DAT的表达以及DA的含量均较成年大鼠显著下降。EGF处理后,老年大鼠自发运动活性、NSDAs中TH和DAT的表达以及黑质(而非纹状体)DA的含量显著上升,但未达到成年大鼠水平。结论:EGF改善老年大鼠自发运动活性衰退伴随黑质DA含量升高。Objective:To evaluate the effect of epidermal growth factor(EGF)on locomotor activity and nigrostriatal dopaminergic system(NSDAs)in aged rats.Methods:The 24-month-old rats were administered with a subcutaneous injection of EGF(0.15 mg/mL)for 7 days.Open field test was used to evaluate locomotor activity.The expression of tyrosine hydroxylase(TH)and dopamine transporter(DAT)in the NSDAs were determined by realtime quantitative PCR and Western blotting analysis.Dopamine(DA)contents in the NSDAs were determined by liquid chromatography coupled with tandem mass spectrometry assay.Results:Aged rats showed significantly reduced behavioral parameters in open field test compared to that in adult rats.The expression of TH and DAT as well as the contents of DA in the NSDAs was significantly decreased in the aged rats compared to those of the adult rats.EGF supplement remarkably ameliorated open field behavioral parameters in aged rats.The expression of TH and DAT in the NSDAs was significantly increased in the aged rats after EGF treatment.The contents of DA in the substantia nigra increased following EGF supplement in the aged rats.Conclusion:EGF significantly ameliorates locomotor activity declines with increased DA contents in the substantia nigra in the aged rats.
