摘 要:目的:通过动物实验,研究异功散早期干预下支气管哮喘模型幼鼠血清短链脂肪酸(SCFAs)及肺组织G蛋白偶联受体43(GPR43)表达的变化,探索异功散防治哮喘的作用机制。方法:7日龄BALB/c雌性幼鼠共32只,随机分为空白组、模型组、中药组(异功散)和益生菌组(酪酸梭菌),以鸡卵清蛋白(OVA)致敏激发法制备哮喘幼鼠模型后,检测其血清SCFAs含量及肺组织GPR43的表达。结果:与空白组比较,模型组血清SCFAs中乙酸浓度升高,丙酸、丁酸浓度降低,但差异均无统计学意义(均P>0.05);肺组织GPR43表达有增高趋势,但差异无统计学意义(P>0.05)。与模型组比较,中药组及益生菌组血清SCFAs中乙酸浓度升高、丁酸浓度下降,差异均无统计学意义(均P>0.05),但血清乙酸相对比值升高,差异有统计学意义(P<0.05),血清丙酸浓度及丙酸相对比值均显著下降(均P<0.05)。中药组肺组织GPR43表达显著高于模型组(P<0.05),但幼鼠血清乙酸浓度及肺组织GPR43表达之间无显著相关(P>0.05)。结论:异功散的早期干预可能影响了哮喘幼鼠肠道菌群的SCFAs代谢,并上调了肺组织GPR43表达,但对这二者的作用并无显著相关性。Objective:To investigate the changes in serum short-chain fatty acids(SCFAs)and lung tissue G-protein-coupled receptor 43(GPR43)expression in a bronchial asthma young mouse model with early intervention of Yigong Powder through animal experiments to explore the mechanism of Yigong Powder in the prevention and treatment of asthma.Methods:Thirty-two 7-day-old female BALB/c mice were randomly divided into a blank group,a model group,a Yigong Powder group,and a probiotics group(Clostridium butyricum).The asthma mouse model was established using ovalbumin(OVA)sensitization and challenge method.Serum levels of SCFAs and lung tissue GPR43 expression were detected.Results:Compared with the blank group,the model group showed increased acetic acid concentration(P>0.05),decreased propionic acid and butyric acid concentrations in serum SCFAs(P>0.05),and an increasing trend of the expression of lung tissue GPR43(P>0.05).Compared with the model group,the Yigong Powder group and the probiotics group showed increased acetic acid concentration(P>0.05),decreased butyric acid concentration in serum SCFAs(P>0.05),increased relative ratio of acetic acid in serum(P<0.05),and reduced serum concentration and relative ratio of propionic acid(P<0.05).Lung tissue GPR43 expression in the Yigong Powder group was significantly higher than that in the model group(P<0.05),but there was no significant correlation between serum acetic acid concentration and lung tissue GPR43 expression in mice(P>0.05).Conclusion:Early intervention with Yigong Powder may influence the SCFAs metabolism of the intestinal flora in asthma mice and upregulate the expression of GPR43 in lung tissue,but there is no significant correlation between these two factors.
