参萸补血方对再生障碍性贫血小鼠骨髓增殖及血液学参数的影响

摘　　要：[目的]观察参萸补血方对再生障碍性贫血(AA)小鼠骨髓细胞学的影响,探讨其治疗AA的确切疗效。[方法]将80只小鼠按随机数字表法分为4组:正常对照组、模型组、参萸补血方组、醋酸泼尼松组,每组20只。除正常对照组外,余组小鼠以60 Co-γ射线照射加腹腔注射环磷酰胺、氯霉素复合方法建立AA模型。造模后参萸补血方组[剂量为16.56 g(/kg·d)]、醋酸泼尼松组[剂量为9.1 mg(/kg·d)]给药连续干预4周,每天灌胃量均为10 ml/kg。光镜下观察骨髓增生程度并计算骨髓非造血细胞百分率(包括淋巴细胞、单核细胞、浆细胞等),血细胞分析仪检测各组小鼠外周血中血液学参数情况。[结果]与正常对照组比较,模型组小鼠外周血中的白细胞(WBC)、血红蛋白(HGB)和血小板(PLT)水平均显著降低,骨髓中非造血细胞的百分率显著升高(P<0.05),骨髓增生低下(P<0.01);与模型组比较,参萸补血方组、醋酸泼尼松组小鼠外周血中WBC、HGB、PLT水平均显著升高,骨髓中非造血细胞百分率显著下降(P<0.05),骨髓增生活跃(P<0.01)。[结论]参萸补血方能够修复AA模型小鼠的骨髓造血功能,促进骨髓增殖,改善外周血液学参数,有效治疗AA。[Objective]To observe the effect of Shenyu Buxue prescription on the myeloid cytology of rats with aplastic anemia(AA)and to explore its exact efficacy.[Methods]Eighty rats were divided randomly into four groups ac⁃cording to the random number table method:normal control group,model group,Shenyu Buxue presciption group,and prednisone acetate group,with 20 rats in each group.Except the normal control group,the rats from the other groups were irradiated with 60 Co-γradiation and intraperitoneal injection of cyclophosphamide and chloramphenicol to estab⁃lish the AA model.After modeling,Shenyu Buxue prescription group with 16.56 g(/kg·d)and prednisone acetate group with 9.1 mg(/kg·d)were administered for 4 weeks,and the daily gavage volume was 10 ml/kg.The degree of bone mar⁃row proliferation was observed under light microscopy and the percentage of non-hematopoietic cells including lympho⁃cytes,monocytes,plasma cells,etc.of bone marrow was calculated,and the hematological parameters in the peripheral blood of each group were detected by the hemocytosis.[Results]Compared with the normal control group,the levels of white blood cells(WBC),hemoglobin(HGB)and platelets(PLT)in the peripheral blood in the model group was signifi⁃cantly reduced,the percentage of non-hematopoietic cells in the bone marrow was significantly increased(P<0.05),and the bone marrow hyperplasia was low(P<0.01).Compared with the model group,the levels of WBC,HGB and PLT in the peripheral blood in the Shenyu Buxue prescription group and the prednisone acetate group increased significant⁃ly,the percentage of non-hematopoietic cells in the bone marrow decreased significantly(P<0.05),and bone marrow hyperplasia was active(P<0.01).[Conclusion]Shenyu Buxue prescription can repair the bone marrow hematopoietic function,promote bone marrow proliferation,improve peripheral hematological parameters,and effectively treat aplastic anemia.
