摘 要:为探讨丹参酚酸B对宫颈癌Hela细胞体外增殖、迁移及凋亡的影响,采用噻唑蓝(MTT)法,测定不同浓度丹参酚酸B对Hela细胞体外活性的影响,通过划痕实验检测Hela细胞的迁移抑制率,利用流式细胞术检测Hela细胞的凋亡率。结果表明,0.02、0.04、0.06、0.08、0.10、0.15、0.20 mg/mL的丹参酚酸B对Hela细胞的生长增殖均有抑制作用,且呈剂量效应关系。丹参酚酸B作用Hela细胞24 h后,Hela细胞迁移抑制率分别为51.20%、67.00%、73.00%、79.10%、89.50%、96.10%、98.70%。丹参酚酸B作用48 h后,Hela细胞凋亡率分别为3.79%、9.64%、14.47%、23.23%、30.18%、37.25%、41.32%。说明丹参酚酸B对Hela细胞生长增殖、迁移均具有抑制作用,能促进Hela细胞的凋亡,且随着浓度增加而增强。To explore the influence of salvianolic acid B on the proliferation,migration and apoptosis of Hela cells in vitro,Hela cell proliferation treated with different concentration of salvianolic acid B is evaluated by MTT assay.The migration inhibition rate is measured by scratch test,and the apoptosis rate is detected by flow cytometry method.The results show that 0.02,0.04,0.06,0.08,0.10.0.15 and 0.20 mg/mL salvianolic acid B can inhibit the growth of Hela cells in a dose-effect relationship.The migration inhibition rates of Hela cells are 51.20%,67.00%,73.00%,79.10%,89.50%,96.10%,and 98.70%respectively.The apoptosis rates of hela cells are 3.79%,9.64%,14.47%,23.23%,30.18%,37.25%,and 41.32%respectively after salvianolic acid B treatment of 48 hours.Therefore,salvianolic acid B can inhibit the growth and migration of Hela cells,and promote the apoptosis of Hela cells,which is enhanced with the increase of concentration.
